摘要
目的探讨还原型谷胱甘肽(GSH)治疗Cr6+致大鼠皮肤损伤的疗效。方法选取30只SD大鼠作为实验动物,将其随机分为2组,即实验组和对照组,每组15只,分别剪去背颈部毛发再用备皮刀剃去绒毛致皮肤完全裸露。皮内注射CrO3(0.1 mol/L)溶液0.5 m L,待1周后皮丘表面角化,用手术刀截去硬痂即形成铬疮。实验组每日涂擦自制GSH凝胶6次,连续1周,对照组同期涂擦自制空白凝胶,每日观察并记录2组大鼠铬疮创面形态、变化和新生肉芽生长情况。结果实验组大鼠痊愈5例,显效3例,总治愈率为53.3%;对照组大鼠痊愈2例,显效1例,总治愈率为20.0%。重复测量数据方差分析结果显示,2组大鼠组别间干预效应、时间效应及组别与时间的交互效应均有统计学意义(P<0.05)。且实验组与对照组大鼠的铬疮创面直径随时间变化的趋势大致相同,实验组大鼠创面直径较对照组呈下降趋势,说明实验组的治疗效果优于对照组。结论 GSH对Cr^(6+)所致的皮肤和肌肉组织损伤有治疗作用,能加快铬疮的治愈和新生组织的生成。
Objective To investigate the effects of reduced glutathione(GSH) in treating Cr6+-induced skin lesions in rats. Methods Thirty SD rats were selected as experimental animals and randomly divided into two groups, i.e., the experimental and control groups, with 15 rats in each group. The rats were clipped on the back and neck and then shaved with a dermatome to expose the skin, and 0.5 m LCr O3(0.1 mol/L) solution was injected intra-dermally. After 1 week, the surface of the skin was keratinized, and the scab was removed with a scalpel to form a chromic sore. The experimental group was given self-made GSH gel 6 times a day for 1 week, while the control group used blank self-made gel at the same time. The morphology and changes of the chrome ulcers as well as the growth of new granulation tissues were observed daily. Results Five cases were healed in the experimental group and another 3 cases were found to be effective, with a total healing rate of 53.3%;2 cases were healed in the control group and 1 case was found to be effective, with a total healing rate of 20.0%. The ANOVA analysis of repeated measurements showed that there were statistically significant effects of intervention, time effect and interaction between groups and time in the two groups(P<0.05). The wound diameter of rats in the experimental group showed a decreasing trend compared with the control group, indicating that the treatment effect of the experimental group was better than the control group. Conclusion GSH has therapeutic effect on skin and muscle tissue injury caused by Cr^(6+) and can accelerate the healing of chrome ulcers and the production of new tissue.
