摘要 目的探讨子痫前期患者中microRNA⁃155⁃5p(miR⁃155⁃5p)通过靶向抑制沉默信息调节因子1(Sirt1)对子痫前期的发病机理的影响。方法收集2018年1月至2018年12月期间,选择本院妇产科收治的子痫前期患者和正常孕妇胎盘样本各25例。采用RT⁃qPCR法检测胎盘中miR⁃155⁃5p和Sirt1表达。另外,在体外过表达miR⁃155⁃5p以及敲除Sirt1检测其对HTR⁃8/SVeno细胞迁移和侵袭的影响。结果子痫前期胎盘中miR⁃155⁃5p表达高于对照组(P<0.01),Sirt1 mRNA和蛋白表达水平在子痫前期患者中均低于对照组(P<0.01)。子痫前期患者的miR⁃155⁃5p表达与Sirt1蛋白表达呈负相关(r=-0.6366,P=0.0169)。在HTR⁃8/SVeno细胞中分别过表达miR⁃155⁃5p和敲除Sirt1均抑制滋养层细胞的侵袭和迁移(P<0.01)。结论miR⁃155⁃5p可能通过抑制Sirt1表达调节滋养层细胞的迁移和侵袭从而影响子痫前期的发生。 Objective To investigate the effect of microRNA-155-5p(miR-155-5p) on the pathogenesis of preeclampsia through targeted inhibition of silent information regulator 1(Sirtl) in patients with preeclampsia.Methods From January 2018 to December 2018,placental samples(n=25) from patients with preeclampsia and normal pregnant women were selected from the obstetrics department of our hospital.RT-qPCR was used to detect miR-155-5p and Sirtl expression in the placenta.In addition,over-expression of miR-155-5p and knockdown of Sirtl was performed to determine its effect on HTR-8/SVeno cells migration and invasion.Results The expression of miR-155-5p in the placenta of the preeclampsia group was higher than that of the control group(P <0.01),and the expression levels of Sirtl mRNA and protein were lower in the patients with preeclampsia than in the control group(P <0.01).There was a negative correlation between miR-155-5p expression and Sirtl protein expression in preeclampsia patients(r=-0.636 6,P=0.016 9).Over-expression of miR-155-5p and knockdown of Sirtl in HTR-8/SVeno cells,respectively,inhibited invasion and migration of trophoblast cells(P <0.01).Conclusion miR-155-5p may regulate the migration and invasion of trophoblast cells by inhibiting Sirtl expression,thus affecting the occurrence of preeclampsia.
