摘要
为大量开发SNP用于构建栽培草莓高质量的遗传图谱,以国产草莓品种红星为母本,日本草莓品种红颜为父本构建F1群体,采用SLAF-seq技术和HighMap软件,对2个亲本和200个F1子代群体进行高通量测序、SNP标记的开发及高密度遗传图谱的构建。通过高通量测序,共获得565919066 reads,平均质量Q30为95.36%,平均GC含量为39.68%,样本GC分布正常。通过生物信息学分析,共获得717881个SLAF标签,2136939个SNP标记,其中有56237个SNP为高质量标记可用于遗传图谱标记的定位和构建。共构建了28个连锁群,总图距为4022.16 cM,该图SNP标记14412个,完整度为99.84%,标记间的平均距离为0.28 cM。通过对遗传图谱单体来源进行评估,结果表明每个个体中较大区段均来源于亲本;通过连锁关系热图对相邻标记间的连锁关系进行分析,结果表明,每个连锁群上的大部分标记顺序与基因组保持一致,标记顺序正确,图谱质量高。这是栽培草莓中首次采用SLAF-seq技术构建的高密度SNP遗传图谱,为栽培草莓的遗传研究和分子标记辅助育种奠定了基础。
For the development of a large number of SNP for the construction of high quality genetic map of cultivated strawberry(Fragaria×ananassa,2 n=8 X=56),Hongxing×Benihoppe crossing populations were sequenced by SLAF-seq that yielded 565919066 reads,with Q30 value of 95.36%and GC content of 39.68%.717881 SLAFs and 2136939 SNPs were identified.Total 56237 SNPs showed high quality for genetic map construction,of which 14412 were successfully mapped to 28 linkage groups spanning a total of 4022.16 cM with an average interval genetic distance of 0.28 cM.It was showed that the greater part of each individual was derived from the parents by evaluating the monomer source of the genetic map.The results of heat map showed that the sequence of most markers on each linkage group was consistent with the genome,the sequence of markers was correct,which could show that the map was of high quality.A high-density genetic map of cultivated strawberry by SLAF-seq could be laying a foundation for genetic research and molecular marker-assisted breeding of cultivated strawberry.
