摘要
以黑木耳(Auricularia heimuer)菌株为试验材料,研究不同温度下黑木耳菌株菌丝生长速率,确定黑木耳培养最适温度;探究不同温度和时间高温胁迫后菌丝恢复生长情况,以确定黑木耳高温胁迫最适条件。结果发现,黑木耳最适培养温度为30℃,高温胁迫最适条件为44℃处理2 h;在此最适高温胁迫条件下研究黑木耳菌丝恢复生长情况,发现不同黑木耳菌株菌丝恢复生长速率、菌丝尖端直径、产色素情况等方面差异显著。本研究为黑木耳耐高温品种选育提供筛选方法,为开展高温胁迫机制研究提供方法学基础。
Auricularia heimuer strains were collected as experimental materials to determin the optimum culture temperature based on mycelia growth rate at different temperatures. The optimum heat stress condition of A. heimuer was determined by the mycelium resumed growth after heat stress at different temperature and time. The results showed that the optimum culture temperature of A. heimuer was 30℃, and the optimum heat stress condition was 44℃ for 2 h, and there were significant differences in mycelium growth rate, diameter of mycelium tip and pigment production among different A. heimuer strains in this condition.This study provides a screening method for breeding high temperature resistant varieties of A. heimuer and a methodological basis for the study of heat stress mechanism.
