银耳多糖提取工艺优化及其对SDS诱导HaCaT细胞损伤的保护作用 被引量:4

Optimization of Extracting Tremella fuciform Polysaccharide and Its Protection Against SDS-induced Damage of HaCaT Cells

摘要 以银耳为原料,采用胶体磨辅助热水浸提工艺提取银耳多糖。以多糖提取率作为评价指标,在单因素试验的基础上,通过正交试验对银耳多糖的提取工艺进行优化,进一步测定了银耳多糖分子量及其对SDS诱导HaCaT细胞损伤的保护作用。研究结果表明,银耳多糖的最佳提取工艺为料液比1∶55,提取温度100℃,提取时间2 h,提取2次。在此条件下,银耳多糖的提取率可以达到(23.04±0.18)%,明显高于常规工业生产工艺。此外,最佳工艺条件下所得主要为100 k Da以下的小分子量银耳多糖,对经由SDS诱导的HaCaT细胞损伤具有明显的保护作用。 The polysaccharide of Tremella fuciformis was extracted from T. fuciformis by colloidal mill assisted with hot water extraction. On the basis of single factor experiment, the extraction process of T. fuciformis polysaccharide was optimized by orthogonal experiment, and the molecular weight of T. fuciformis polysaccharide and its protection against SDS-induced damage of HaCaT cells were further determined. The results showed that the best extraction technology of T. fuciformis polysaccharide was material liquid ratio 1 ∶ 55, extraction temperature 100℃, extraction time 2 h, extraction twice. Under this condition, the extraction rate of T. fuciformis polysaccharide could reach(23.04 ± 0.18)%, which was significantly higher than the conventional industrial production technology. In addition, under the optimal conditions, the main small molecular weight T. fuciformis polysaccharide under 100 kDa was obtained. The experiment confirmed that the T. fuciformis polysaccharide had the protection against SDS-induced damage of HaCaT cells.
