组织型转谷氨酰胺酶与转化生长因子β1在幼鼠肾间质损伤中的表达及意义

摘要 目的探讨组织型转谷氨酰胺酶和转化生长因子β1在幼鼠肾间质损伤中的表达及意义.方法60只幼鼠按照随机数字表法分为假手术组、模型组和试验组共3组,每组20只.选择试验组和模型组幼鼠进行单侧输尿管结扎模型的建立,试验组从建模后第2d起灌胃给予苯那普利和氯沙坦.于造模后第3 d、7 d、14 d、28 d,分别处死5只大鼠,取肾组织,采用Masson染色检测3组肾组织常规病理变化,检测3组组织型转谷氨酰胺酶和转化生长因子β1表达水平.结果模型组、试验组肾间质纤维组织相对面积显著高于假手术组(P<0.05),模型组显著高于试验组(P<0.05).3组转化生长因子β1 mRNA吸光度值比较差异有统计学意义(P<0.05),模型组、干预组吸光度值随时间增长逐渐增加,在操作第14 d时达到高峰.模型组、试验组转化生长因子β1 mRNA表达水平均显著高于假手术组(P<0.05),试验组显著低于模型组(P<0.05).结论在肾间质纤维化过程中,组织型转谷氨酰胺酶和转化生长因子β1互相作用,并且参与其中,苯那普利可以调节组织型转谷氨酰胺酶和转化生长因子β1的表达水平,可能会成为潜在有效靶点. Objective To investigate the expression and significance of tissue transglutaminase(tTG)and transforming growth factorβ1(TGF-β1)in renal interstitial injury in youngrats.Methods 60 youngrats were randomly divided into three groups:sham operation group,model group and experimental group,20 cases each.The unilateral ureteral ligation model was established in the experimental group and the model group.The experimental group was given benazepril and losartan from the 2nd day after modeling.On the 3rd,7th,14th and 28th day after model establishment,5 rats were sacrificed and kidney tissues were taken.The routine pathological changes of renal tissues were detected by M asson staining.The tTG and TGF-β1 expression levels of the three groups were examined.Results The relative area of renal interstitial fibrous tissue in model group and experimental group was significantly higher than that in sham operation group(P<0.05),the model group was significantly higher than the experimental group(P<0.05).There were significant differences in the absorbance values of TGF-β1 mRNA among the three groups(P<0.05).The absorbance values of the model group and the intervention group gradually increased w ith time,and reached a peak at 14 d.The expression of TGF-β1 mRN A in the model group and the experimental group was significantly higher than that in the sham operation group(P<0.05).The experimental group was significantly lower than the model group(P<0.05).Conclusions In the process of renal interstitial fibro-sis,tTG and TGF-β1 interact and participate in it.And benazepril can regulate the expression of tTG and TGF-β1,and may become a potentially effective target.
