摘要
利用可逆加成-断裂链转移(RAFT)聚合与开环聚合相结合的方法,合成了4-硝基苯基丙烯酸酯链段为反应位点的嵌段共聚物,利用点击化学原理,选择1,8-辛二胺作为交联剂进行交联反应,制备嵌段间面交联胶束体系。利用核磁共振波谱仪、凝胶渗透色谱、透射电镜等方法对聚合物相对分子质量及其分布、交联胶束形态进行表征。结果表明,成功合成了嵌段间面交联胶束体系。药物的搭载与释放性能测定及细胞毒性试验分析结果表明,该胶束系统生物相容性好,稳定性高,对阿苯哒唑药物搭载能力可达45%,在酸性环境中,药物缓释过程具有pH响应性。
Block copolymers containing 4-nitrophenylacrylate fragments were synthesized by reversible addition-fragmentation chain transfer(RAFT)polymerization combined with ring-opening polymerization.The nexus-crosslinked micelles were formed by using the principle of click chemistry,N-nitrophenyl acrylate(NPA)as functional group and 1,8-diaminooctane as crosslinker.The molecular weight,molecular weight distribution and morphology of crosslinked micelles were characterized by NMR,SEC and TEM.The results of drug loading,release performance and cytotoxicity test show that the micelle system has a good biocompatibility and high stability.The drug loading capacity of albendazole is up to 45%.In acidic environment,the drug release process is pH responsive.
