蒙药蓝刺头对绝经后骨质疏松症大鼠瘦素、β_(2)肾上腺素受体的影响

Effects of Mongolian Medicine Lancitou on Leptin and β_(2) Adrenaline Receptors of PMOP Rats

目的:从预防骨丢失和促进骨再生两方面研究蒙药蓝刺头对绝经后骨质疏松症的防治作用,阐述其促进抑制破骨作用的信号通路。方法:通过去卵巢复制绝经后骨质疏松症模型;用蒙药蓝刺头、强骨胶囊和己烯雌酚分别灌胃模型大鼠90天,观察比较各组大鼠骨密度(bone mineral density,BMD)、骨小梁体积/组织体积(bone trabecula volume/tissue volume,BV/TV)、子宫重量/体质量(uterus weight/body weight,UW/BW)及瘦素(leptin,LEP)、股骨β2肾上腺素能受体(β_(2)-adrenergic receptor,ADRβ_(2)R)的表达。结果:与模型组比较,蒙药蓝刺头能从骨密度方面有效防治绝经后骨质疏松症模型大鼠疏松骨质,能降低绝经后骨质疏松症模型大鼠血清LEP水平(P<0.01)及ADRβ_(2)R的表达(P<0.05)。结论:蒙药蓝刺头具有抑制骨吸收、促进骨形成的作用,其防治骨质疏松的作用机制涉及LEP/ADRβ_(2)R信号通路,这为基于蒙医“肾主骨,治以补暖补精”理论防治骨质疏松提供了实验依据。

Objective:To study the effects of mongolian medicine Lancitou(echinops sphaerocephalus)on the prevention of postmenopausal osteoporosis(PMOP)from preventing bone loss and promoting bone regeneration,and elucidate the signaling pathway of promoting and inhibiting bone-destroying effects.Methods:PMOP models were established by ovariectomizing;rat models were drenched with mongolian medicine Lancitou,Qianggu capsules and diethylstilbestrol for 90 days,to observe and compare bone mineral density(BMD)in different groups,bone trabecula volume/tissue volume(BV/TV),uterine weight/body weight(UW/BW),leptin(LEP)and the expressions of femoral β_(2) adrenergic receptors(ADRβ_(2)R).Results:Compared with the model group,mongolian medicine Lancitou could effectively prevent osteoporosis of PMOP rats from BMD and reduce LEP levels(P<0.01),and lower the epressions of ADRβ_(2)R(P<0.05).Conclusion:Mongolian medicine Lancitou owns the effects of inhibiting bone resorption and promoting bone regeneration,and its mechanism of preventing osteoporosis involves LEP/ADRβ_(2)R signaling pathway,and it could provide experimental reference for the prevention and treatment of PMOP by classical treatment of mongolian medicine.
