鸽圆环病毒新疆株全基因组克隆与序列分析 被引量:3

Cloning and analysis of the complete genomes of pigeon circovirus from Xinjiang

摘要 采用PCR扩增与基因序列测定方法从病死鸽肝组织扩增获得鸽圆环病毒(Pigeon circovirus,PiCV)新疆株(命名为XJ1株)全基因组序列,并对其基因组组成、结构及遗传进化关系进行分析,结果显示,XJ1株基因组全长为2031 bp,存在ORF V1(41~994 nt)、ORF C1(1166~1981 nt)、ORF C2(410~790 nt)、ORF C3(2~292 nt)和ORF C4(739~984 nt)5个阅读框,5’端和3’端基因间隔区长度分别为90 nt和171 nt。XJ1与已报道的24株鸽圆环病毒全基因组序列同源性为87.2%~94.9%,其中与GF17和GH1834的同源性最高(均为94.9%),基于基因遗传进化树显示,XJ1株与Cap蛋白起始密码子"ATG"分支中GF82、JF007及GH1834亲缘关系最近。鸽圆环病毒新疆株全基因组序列的分析为疫苗开发提供资料参考。 The whole genome sequence of the Xinjiang strain(named XJ1 strain)was amplified from the liver tissue of dead pigeon by PCR method and gene sequencing,and then the nucleotide composition,structure and genetic evolution of the XJ1 strain were also analyzed.The results showed that the genome length of XJ1 strain was 2031 bp,there were five reading frames(ORFs)including ORF V1(41-994 nt),ORF C1(1166-1981 nt),ORF C2(410-790 nt),ORF C3(2-292 nt)and ORF C4(739-984 nt).The length of the 5’-and 3’-terminal gene spacer was 90 nt and 171 nt,respectively.The genome sequences homology between XJ1 strain and other 24 published strains was 87.2%-94.9%,among them,it has the highest homology with GF17 and GH1834(94.9%).The genetic evolution tree based on Cap gene showed that XJ1 strain had most closely related to strains GF82,JF007 and GH1834 initiation codon"ATG"branch.This is the first time to determine the whole genome sequence of pigeon circovirus in Xinjiang.The analysis of the complete genomes of pigeon circovirus from Xinjiang provides references for vaccine developme.
