摘要
为了解H5N2亚型禽流感病毒(Avian influenza virus,AIV)的血凝素蛋白(Hemag-glutinin,HA)系统进化特征,借助NCBI influenza virus resource数据库,下载禽源A型流感病毒(Influenza A Virus,IAV)H5N2亚型HA核酸序列,分析毒株的宿主、分离时间等特征,并构建HA蛋白的遗传进化树,分析其系统发生。结果显示:NCBI数据库收录的H5N2 AIV的HA核酸序列1272条,序列在NCBI发布时间集中于2005—2007年、2011年、2014—2016年;582条序列为野禽来源,宿主主要分布于雁形目、鸡形目和鸻形目的野禽。系统发生分析结果表明:H5N2 AIV的HA蛋白可聚类为亚欧谱系及北美谱系两大分支,前者可分为三个亚系,后者可分为两个亚系。研究结果呈现了H5N2 AIV HA基因在时间及空间上的进化特征,为H5N2AIV引起的疫病预警和防控提供理论基础。
The aim of the present study was to systematically present the evolutionary characteristics of hemagglutinin(HA)gene from H5 N2 subtype avian influenza virus(AIV).The HA sequences of avian-origin H5 N2 subtype influenza A virus(IAV)were downloaded from NCBI influenza virus resource database and analyzed the host and isolation time of the strains.Phylogenetic tree of HA protein was built by the corresponding functional module from the database.The results showed that in total 1272 HA nucleic acid sequences of H5 N2 AIV were collected in this study,the sequences were published mainly in 2005 to 2007,2011 and 2014 to 2016.The hosts of 582 sequences were wild birds,principally belonging to Anatidae,Galliformes and Charadriiformes.Phylogenetic analysis showed that HA protein of H5 N2 AIV could be clustered into two branches:Eurasian lineage and North American lineage.Basing on the isolating time and the host characteristics,the former one could be divided into three sub-lineages and the latter could be divided into two sub-lineages.The results provided a theoretical basis for early warning and the prevention and control measures of avian influenza caused by H5 N2 AIV.
