响应面优化复方黄芪饲料添加剂提取工艺与体外抗氧化作用研究 被引量:7

摘要 试验考察液料比、提取时间和提取次数对复方黄芪饲料添加剂中多糖和总皂苷得率的影响,通过响应面进行优化,同时进行体外抗氧化性研究。结果表明,对多糖和总皂苷得率影响顺序为:液料比>提取次数>提取时间。适宜的提取工艺:液料比12 mL/g、提取2次、提取100 min。在此条件下,多糖和总皂苷得率分别为32.10%和7.08%。复方黄芪多糖和总皂苷提取工艺稳定可行、得率较高。复方黄芪对1,1-二苯基-2-三硝基苯肼自由基(·DPPH)和羟自由基(·OH)有一定的清除能力,其清除能力对浓度有一定依赖性;复方黄芪有一定的总抗氧化能力。 The reflux extraction process of polysaccharide and total saponins from Radix astragali compound was optimized by response surface methodology. The liquid-solid ratio, extraction time and extraction times were investigated respectively. The antioxidant properties were also studied in vitro. The effect of liquid to material ratio, extraction time and extraction times on the extraction of polysaccharide and total saponins was studied by the single factor experiment. The order of influence on the yield of polysaccharides and total saponins was liquid to material ratio>extraction times>extraction time. The suitable extracting condition of polysaccharide and total saponins from Astragalus membranaceus compound were liquid to material ratio of 12 mL/g, extraction time twice, and extraction time 100 min. The polysaccharide and total saponin extraction rate of Astragalus membranaceus compound were 32.10% and 7.08%. The method is showed to be stable, feasible and high yield. The antioxidant activity of compound feed additive of Astragalus membranaceus in vitro was investigated by VC. The results showed that Astragalus compound had certain scavenging ability to·DPPH and·OH within a certain concentration range, indicating that Astragalus compound had certain total antioxidant ability.
