Notch1与Wnt10b在人骨肉瘤组织中的表达及临床意义

摘要 目的探讨人骨肉瘤与骨软骨瘤组织中Notch1、Wnt10b表达的规律。方法采用免疫组织化学染色方法检测骨肉瘤组(观察组)46例与骨软骨瘤组(对照组)32例组织标本中Notch1、Wnt10b蛋白表达情况,分析其与临床病理及预后的关系。结果 Notch1、Wnt10b在人骨肉瘤中表达的阳性率分别为是87.0%(40/46)、65.2%(30/46),明显高于骨软骨瘤6.3%(2/32)、34.4%(11/32),差异均有统计学意义(P<0.05)。根据人骨肉瘤病Ennecking的分期,发现骨肉瘤细胞Ⅱ、Ⅲ期Notch1、Wnt10b阳性表达率在明显高于Ⅰ期(P<0.05);比较肿瘤转移组与非转移组,发现Notch1、Wnt10b在转移组阳性表达率分别为94.1%(16/17)、88.2%(15/17),非转移组阳性率分别为13.8%(4/29)、20.7%(6/29),差异有统计学意义(P<0.05)。结论 Notch1、Wnt10b在骨肉瘤中呈现高表达,且与肿瘤恶性程度及肿瘤转移关系密切,检测Notch1与Wnt10b对骨肉瘤进展及预后的判断具有重要指导意义。 Objective To explore the expression of Notch1 and Wnt10 b in human osteosarcoma and osteochondroma.Methods Immunohistochemical staining was used to detect the expression of Notch1 and Wnt10 b protein in osteosarcoma experimental group(46 cases) and osteochondroma control group(32 cases). The relationship between the expression of Notch1 and Wnt10 b protein and clinicopathology and prognosis was analyzed.Results The positive rates of Notch1 and Wnt10 b expression in human osteosarcoma were 87.0%(40/46) and 65.2%(30/46), respectively, which were significantly higher than the expression of osteochondroma 6.3%(2/32) and 34.4%(11/32), the differences were statistically significant(P<0.05). According to the stage of human osteosarcoma Ennecking, it was found that the positive expression rates of Notch1 and Wnt10 b in osteosarcoma cells were significantly higher in phase Ⅱ and Ⅲ than phase Ⅰ(P<0.05). Comparing the tumor metastasis group and the non-metastatic group, it was found that the positive expression rates of Notch1 and Wnt10 b in the metastasis group were 94.1%(16/17) and 88.2%(15/17), respectively, and the positive rates in the non-metastatic group were 13.8%(4/29) and 20.7%(6/29), the difference in positive rate expression between the two groups was statistically significant(P<0.05).Conclusion Notch1 and Wnt10 b are highly expressed in osteosarcoma, and are closely related to tumor malignancy and tumor metastasis. The detection of Notch1 and Wnt10 b has important guiding significance for the progression and prognosis of osteosarcoma.
