摘要
建立了基于氰化高铁血红蛋白定量检测人全血中血红蛋白的方法。该方法在0.3~1.8g/L范围内,线性相关系数为0.9994;日内精密度、日间精密度和重复性实验相对标准偏差分别为0.93%、1.14%和1.97%。用所建立的方法对Hb标准品进行了检测,检测值为116.67g/kg,符合Hb标准品证书值(119.7g/kg,不确定度为3.7g/kg,k=2)。本方法参与了由德国联邦物理技术研究院主导的血红蛋白国际比对,测试结果显示,样品浓度为124.36g/kg,扩展不确定度为2.14g/kg(k=2),比对结果与参考值等效一致。此方法操作简单,测量精确度高,重复性好,可作为全血中Hb标准物质的定值方法,为全血中Hb的日常检测提供参考。
A method for quantitative detection of hemoglobin(Hb)in whole human blood based on HiCN was established.The linear correlation was observed within the range of 0.3~1.8g/L(R2=0.9994).The relative standard deviations of intra-,inter-day,and repeatability were 0.93%,1.14%,and 1.97%,respectively.Hb standard sample was tested by this established method,and the result showed that the concentration of the Hb standard sample was 116.67g/kg,which was in line with the certificate value of Hb standard sample(119.7g/kg,uncertainty 3.7g/kg,k=2).The method was used for the Hb international comparison,led by Physikalisch-Technische Bundesanstalt.The results showed that the sample concentration was 124.36g/kg,and the expanded uncertainty was 2.14g/kg(k=2),which was consistent with the reference value.The quantitative optical detection method of Hb based on HiCN is simple,accurate,and repeatable.It can be used as a method to determine Hb standard substance in whole blood and provides a reference for daily monitoring of Hb in whole blood.
