高表达MMP14与肾透明细胞癌预后及影响机制研究

Correlation between Highly Expressed MMP14 and the Prognosis of Kidney Renal Clear Cell Carcinoma

目的:研究MMP14表达水平与肾透明细胞癌(KIRC)预后的相关性。方法:从TCGA、GTEx和GENT数据库获得mRNA表达谱和临床数据,使用HPA数据库调取MMP14蛋白表达的免疫组化图像进行验证,根据MMP14在肾癌中的不同mRNA表达量进行功能和信号通路富集。结果:与正常肾脏比较,MMP14的mRNA和蛋白表达在KIRC患者中均显著增高,生存曲线提示MMP14上调与KIRC的不良预后显著相关(P<0.05),功能和信号通道富集分析提示高表达MMP14在KIRC中与DNA损伤和WNT信号通路有关。结论:MMP14在KIRC中可能与不良预后相关,并可能是KIRC的潜在预后标记物。

Objective:To study the association between MMP14 expression level and the prognosis in kidney renal clear cell carcinoma(KIRC).Method:The mRNA expression profile and clinical data were obtained from The Cancer Genome Atlas(TCGA),Genotype-Tissue Expression(GTEx),and Gene Expression across Normal and Tumor tissue(GENT)database.The protein expression was investigated from immunohistochemistry images using the Human Protein Atlas(HPA)database.The enrichment analyses of functional and signaling pathways based on the different MMP14 mRNA expression level were performed using gene set enrichment analysis(GESA).Result:Compared with normal kidney,the mRNA and protein expression of MMP14 were significantly higher in patient with KIRC(P<0.05).The Kaplan-Meier survival analysis revealed that up-regulation of MMP14 was significantly associated with poor survival in KIRC(P<0.05).The functional and signaling pathways enrichment analyses indicated the high expression of MMP14 was related to DNA damage and WNT signaling pathway in KIRC.Conclusions:MMP14 may be associated with the poor prognosis in KIRC,and may be a potential prognostic marker for KIRC.
