基于“阳中求阴”理论研究左归丸对肾阴虚证能量代谢的影响 被引量:5

Effect of Zuogui Pill on Energy Metabolism of Kidney-Yin Deficiency Syndrome Based on"Reinforcing Yin from Yang"Theory

摘要 目的:通过观察左归丸及其拆方对肾阴虚大鼠肝线粒体能量代谢的影响,探讨左归丸“阳中求阴”治疗肾阴虚证的作用机理。方法:采用灌胃甲状腺素片与利血平混合悬浮液制备肾阴虚证模型,并随机分为模型组、左归丸组、滋阴组和补阳组。检测大鼠肝组织线粒体中SDH、Na^(+)-K^(+)-ATPase、CCO、LDH活性及ATP含量变化,电镜下观察肝组织中线粒体超微结构的变化。结果:与对照组比较,模型组大鼠肝线粒体SDH、Na^(+)-K^(+)-ATPase、CCO、LDH活性及ATP含量均显著下降(P<0.01),肝线粒体超微结构损伤严重;与模型组比较,左归丸组上述酶的活性及ATP含量均显著升高(P<0.05或P<0.01),肝线粒体超微结构的病理变化明显改善。结论:干预肝线粒体能量代谢可能是左归丸“阳中求阴”治疗肾阴虚证的作用机理,进一步体现左归丸“阳中求阴”的配伍意义。 Objective:To observe the effect of Zuogui energy metabolism of liver mitochondrial in Kidney-yin deficiency rats,and to explore the mechanism of Zuogui Pill in treating Kidney-yin deficiency syndrome with“reinforcing yin from yang”.Methods:A mixed suspension of Thyroxine and Reserpine was used to prepare the rat model of Kidney-yin deficiency syndrome,the successful models were randomly divided into the model group,the Zuogui Pill group,the Ziyin group and the Buyang group.The activities of SDH,Na^(+)-K^(+)-ATPase,CCO and LDH,as well as the content of ATP were detected.The ultrastructure of mitochondria in liver tissue was observed under transmission electron microscope.Results:Compared to those in the control group,the activities of SDH,Na^(+)-K^(+)-ATPase,CCO and LDH and ATP content of liver mitochondria were significantly decreased in the model group(P<0.01),and the ultrastructural of liver mitochondria was severely damaged.The activity of the above enzymes and ATP content were significantly increased in Zuogui Pill group compared to those in the model group(P<0.05,P<0.01);the pathological changes of liver mitochondrial ultrastructure were significantly improved.Conclusion:Intervention of energy dysfunction of liver mitochondria may be the mechanism of Zuogui Pill in treating Kidney-yin deficiency with the theory of“reinforcing yin from yang”,which further reflects the compatibility significance of“reinforcing yin from yang”in Zuogui Pill.
