霍山石斛不同提取物对肾阴虚证和肾阳虚证小鼠温度趋向性、环核苷酸及甲状腺激素的影响

Effects of different extract of Dendrobium huoshanense on temperature tendency,cyclic nucleotides and thyroid hormones in mice with kidney yin deficiency syndrome and kidney yang deficiency syndrome

目的:研究霍山石斛不同提取物对肾阴虚证、肾阳虚证小鼠温度趋向性、环核苷酸及甲状腺素水平的影响,探讨霍山石斛寒凉药性的物质基础。方法:180只小鼠随机分为15组,每组12只。连续14d分别灌服甲状腺素(160mg/kg)、氢化可的松溶液(25mg/kg)制备肾阴虚、肾阳虚动物模型,观察各组小鼠的一般状态、体质量和温度趋向性,检测血清cAMP、cGMP、T3、T4水平,计算cAMP/cGMP比值。结果:与肾阴虚模型组比较,实验第14天,霍山石斛水提物及醇提物可增加小鼠的体质量和热区停留比例(P<0.05,P<0.01),降低cAMP含量和T4的含量(P<0.05,P<0.01),霍山石斛水提物及醇提物高剂量组能降低cAMP/cGMP值(P<0.01),降低T3含量(P<0.05,P<0.01);与肾阳虚模型组比较,实验第14天,霍山石斛水提物高剂量可增加小鼠的体质量(P<0.01),降低其热区停留比例(P<0.05),升高cAMP值、cAMP/cGMP值及T3、T4含量(P<0.05,P<0.01)。结论:霍山石斛水提物对肾阴虚证和肾阳虚证的机体状态、环核苷酸水平及甲状腺激素水平均有调节作用,表现出平性药的药性特点;霍山石斛醇提物仅对肾阴虚证有调节作用,表现出寒凉药性;由此推断,霍山石斛醇提物是其寒凉药性的物质基础,水提物和醇提物综合作用使霍山石斛显现微寒之性。

Objective:Studying the effects of different extracts of Dendrobium huoshanense in mice with kidney yin deficiency syndrome and kidney yang deficiency syndrome to explore the material basis of cold cooling properties of Dendrobium huoshanense.Methods:A total of 180 mice were divided into 15 groups,12 mice in each group.The animal models of kidney yin deficiency and kidney yang deficiency were established by administration of thyroxine(160 mg/kg)and hydrocortisone solution(25 mg/kg)for 14 days.The general state,body weight and temperature trend of mice in each group were observed.Detecting serum cAMP,cGMP,T3,T4 levels,and calculating cAMP/cGMP ratio.Results:Comparison with kidney yin deficiency model group,on the 14 th day of the experiment,the water extracts of Dendrobium huoshanense and alcohol extracts of Dendrobium huoshanense can increase the body weight of mice and the retention ratio of hot zone in mice(P<0.05,P<0.01),and reduce the content of cAMP and T4(P<0.05,P<0.01).The high-dose group of water extract of Dendrobium huoshanense and alcohol extract of Dendrobium huoshanense can reduce the value of cAMP/cGMP(P<0.01)and decrease the content of T3(P<0.05,P<0.01).Comparison with kidney-yang deficiency model,on the 14 th day of the experiment,the high dose of water extract of Dendrobium huoshanense can increase the body mass(P<0.01)and decrease retention ratio of hot zone in mice(P<0.05),and rise the cAMP,cAMP/cGMP,T3 and T4 contents(P<0.05,P<0.01).Conclusion:The water extract of Dendrobium huoshanense can regulate the body state,cyclic nucleotides level and thyroid hormone level of yin deficiency syndrome and yang deficiency syndrome,showing the characteristics of flat drugs.The alcohol extract of Dendrobium huoshanense only has a regulating effect on kidney-yin deficiency syndrome,and shows cold and cool properties.It is inferred that the alcohol extract of Dendrobium huoshanense is the material basis of its cold and cool properties;and the combined action of water extract and alcohol extract makes Dendrobium huoshanense show cold relaxation and thus has the property of slight cold.
