加味四妙勇安油治疗大鼠急性放射性皮肤损伤机制 被引量:6

Mechanism of modified Simiao Yongan Oil in acute-radioactive induced skin reaction rats

摘要 目的:探讨加味四妙勇安油在治疗放疗导致的急性放射性皮肤损伤过程中的抗炎、保护胶原纤维及促血管新生作用。方法:将75只大鼠随机分为对照组、B12组、四妙勇安组,每组25只,暴露于直线加速器下,经6MeV-X电子射线照后产生10%Ⅱ度放射性皮炎模型,观察照射野皮肤淋巴细胞浸润、胶原纤维排列和完整性,血管内皮生长因子(VEGF)、血管内皮生长因子受体(VEGFR)、血小板源性生长因子-α(PDGF-α)、血小板源性生长因子-α受体(PDGF-αR)的表达。结果:形态学染色显示,皮损急性期,四妙勇安组皮损区淋巴细胞浸润较其他组明显减少,胶原纤维排列较其他组整齐,外形更加完整。免疫组化显示,四妙勇安组VEGF、VEGFR、PDGF-α、PDGF-αR反应点均多于其他两组,其中PDGF-αR、VEGF表达在第15天之后差异显著,PDGF-α在第30天之后差异显著。VEGFR表达前期显著增加,第45天开始四妙勇安组与B12组的差异逐渐消失,以上差异具有统计学意义(P<0.05)。结论:加味四妙勇安油在治疗放疗导致的急性放射性皮肤损伤时能有效减轻炎症反应,保护胶原纤维,促进血管及成纤维细胞生成,加快损伤的愈合。 Objective:To investigate the possible mechanism of anti-inflammatory,collagen protective and angiogenesis effects of modified Simiao Yongan Oil in acute-radioactive induced skin reaction.Methods:Seventy-five rats were randomly divided into control group,B12 group and SMYA group,with 25 rats in each group.After exposure to linear accelerator,10%II degree radiation dermatitis model was produced.The infiltration of skin lymphocytes,the arrangement and integrity of collagen fibers,vascular endothelial growth factor(VEGF),vascular endothelial growth factor receptor(VEGFR),platelet-derived growth factor-α(PDGF-α)and platelet-derived growth factor-αreceptor(PDGF-αR)were observed.Results:HE staining and Sirius red staining showed that lymphocyte infiltration in skin lesions in SMYA group was significantly reduced and the collagenous fibers in SMYA group were more orderly and more complete than those in other groups.Immunohistochemistry showed that the reaction points of VEGF,VEGFR,PDGF-αand PDGF-αR in SMYA group were more than those in the other two groups,the expressions of PDGF-αR and VEGF were significantly different after the 15 th day,and PDGF-αwas significantly different after the 30 th day.The expression of VEGFR increased significantly in the early stage,and the difference between SMYA group and B12 group gradually disappeared from the 45 th day,with statistical significance(P<0.05).Conclusion:Modified Simiao Yongan Oil can effectively reduce the lymphocyte infiltration in the damaged area of skin tissue,protect the integrity and organization of collagen fibers,and it can also promote vascular and fibroblast to generate,accelerate wound healing.
