摘要
建立了一种快速测定巴戟天中多糖含量的方法:样品经液氮粉碎处理后,先用80%乙醇为溶剂超声提取除去单糖和低聚糖,过滤后残渣再用纯水为溶剂在超声下提取多糖,最后用苯酚硫酸法显色测定多糖含量。实验探讨了液氮粉碎次数及超声提取温度和时间对测定结果的影响,并与传统的加热回流提取法进行了对比研究,建立了两提取方法间的结果换算公式。结果表明,本法操作简单、快速,时间上缩减了约3/4,其样品检测结果经公式换算后与加热回流法的相对误差小于10%。本法可满足企业快速检测产品质量的要求,具有较高的实用价值。
A method is established for rapid determination of the contents of polysaccharides in Morindae Officinalis Radix.After being grinded with liquid nitrogen,monosaccharides and oligosaccharides are extracted using 80%ethanol as solvent with ultrasonic extraction.The polysaccharides are extracted with water as solvent under ultrasound from the filter residue and determined by the method of phenol-sulfuric acid.In this paper,the effects of grinding times and ultrasonic extraction temperature and time on the determination results are discussed.This method is compared with the traditional method,in which polysaccharides are extracted with heating reflux,and a conversion formula about experimental results of two methods is established.The results show that this method is simple and fast and can save time by 3/4.The relative errors between the conversion results of this method of formula and the results of traditional method are less than 10%.This method can rapidly determine the quality of product in enterprise,and has higher application value.
