垂体肿瘤转化基因在膀胱癌组织中的表达及其对T24细胞的影响

摘要 目的探讨垂体肿瘤转化基因(PTTG)在膀胱癌组织中的表达水平及其对T24细胞恶性生物学行为的影响。方法应用免疫组化法检测膀胱癌患者膀胱癌组织与癌旁组织中PTTG蛋白表达量,比较不同年龄、性别、病灶直径、淋巴结转移、临床分期患者膀胱癌组织中PTTG蛋白表达水平,应用Spearman分析PTTG与各指标的相关性。将PTTG siRNA与空载siRNA分别转染T24细胞,应用Western blot法检测PTTG siRNA组、空白对照组和空载siRNA组T24细胞中PTTG蛋白表达水平,MTT法检测细胞增殖情况,流式细胞术检测细胞凋亡情况,Transwell实验检测细胞侵袭能力。结果膀胱癌组织中PTTG蛋白表达水平明显高于癌旁组织(P <0.05)。不同年龄、性别、病灶直径患者膀胱癌组织中PTTG蛋白表达水平比较,差异无显著意义(P> 0.05);淋巴结转移者PTTG蛋白表达高于无淋巴结转移者,不同临床分期、分化程度患者PTTG蛋白表达有显著差异(P <0.05);PTTG蛋白表达水平与淋巴结转移、临床分期呈正相关,与分化程度呈负相关(P <0.05)。PTTG siRNA组PTTG蛋白表达水平低于空白对照组和空载siRNA组(P <0.05)。siRNA转染2、3、4 d,PTTG siRNA组细胞增殖抑制率高于空载siRNA组(P <0.05)。PTTG siRNA组T24细胞凋亡率高于空白对照组和空载siRNA组,穿matrigel胶的细胞数低于空白对照组和空载siRNA组(P <0.05)。结论 PTTG蛋白在膀胱癌组织中高表达,与淋巴结转移、临床分期正相关,沉默PTTG可抑制T24细胞增殖和侵袭,促进T24细胞凋亡。 AIM To explore the expression level of pituitary tumor transforming gene(PTTG) in bladder cancer and its effect on the malignant biological behavior of T24 cells. METHODS The expression of PTTG protein in bladder cancer tissues and adjacent tissues of cancer was detected by immunohistochemistry. The expression level of PTTG protein in bladder cancer tissues of patients with different ages, genders, lesion diameters, lymph node metastasis and clinical stages were compared, and the correlation between PTTG protein level and various indicators was analyzed by Spearman. PTTG siRNA and empty siRNA were respectively transfected into T24 cells.Western blot was used to detect the expression level of PTTG protein in T24 cells in PTTG siRNA group, blank control group and empty siRNA group. Cell proliferation was detected by MTT assay, apoptosis was detected by flow cytometry, and cell invasion was detected by Transwell assay. RESULTS The expression level of PTTG protein in bladder cancer tissue was significantly higher than that in adjacent tissues(P < 0.05). There was no significant difference in the expression level of PTTG protein in bladder cancer tissues of patients with different ages, genders and lesion diameters(P > 0.05). The expression of PTTG protein in patients with lymph node metastasis was higher than that in those without lymph node metastasis, and there were significant differences in PTTG protein expression in patients with different clinical stages and degrees of differentiation(P < 0.05). The expression level of PTTG protein was positively correlated with lymph node metastasis and clinical stage, and negatively correlated with the degree of differentiation(P < 0.05). The expression level of PTTG protein in the PTTG siRNA group was lower than that of the blank control group and the empty siRNA group(P < 0.05). The inhibition rate of cell proliferation in the PTTG siRNA group was higher than that in the empty siRNA group on 2, 3, and 4 days after siRNA transfection(P < 0.05). The apoptosis rate of T24 cells in the PTTG siRNA group was higher than that of the blank control group and the empty siRNA group, and the number of cells penetrating the matrigel was lower than the blank control group and the empty siRNA group(P < 0.05). CONCLUSION PTTG protein is highly expressed in bladder cancer tissues and is positively correlated with lymph node metastasis and clinical staging. Silencing PTTG can inhibit the proliferation and invasion of T24 cells and promote the apoptosis of T24 cells.
