在线提取HPLC-ABTS联用快速分析枳椇子中抗氧化成分 被引量:4

Rapid analysis of antioxidant in Hoveniae Semen by online extraction HPLC-ABTS

摘要 目的:建立在线样品提取HPLC-ABTS联用的方法快速分析枳椇子中的抗氧化活性成分。方法:枳椇子样品与硅藻土混合,置于在线提取罐中,用流动相进行在线提取。提取液采用核壳反相色谱柱分离,VWD检测器350 nm检测提取液化合物色谱图。分离后的化合物与ABTS溶液混合反应,用DAD检测器在750 nm检测其抗氧化活性化合物色谱图。结果:新建立的分析方法在15 min内完成了枳椇子样品的提取、分离和抗氧化活性检测,通过质谱数据鉴定了10个化合物,其中4个成分具有抗氧化活性,均为黄酮类成分,二氢杨梅素为主要抗氧化活性成分。结论:新建立的分析方法快速、稳定、环保,为枳椇子的化学研究及产品开发提供了依据。 Objective:To develop on-line extraction HPLC-ABTS method for rapid screening antioxidant in Hoveniae Semen.Methods:The sample powder was mixed with diatomite,put into the on-line extraction tank,and extracted by mobile phase on-line.HPLC separation was performed on a poroshell reverse phase column,and the chromatogram of Hoveniae Semen was detected at 350 nm by VWD.After the HPLC separation,the analytes reacted with ABTS solution,and the chromatogram of antioxidant compounds was detected at 750 nm by DAD.Results:The results showed that the new method completed the sample extraction,separation and antioxidant screening within 15 min.10 components were identified and 4 flavonoids showed antioxidant activity with dihydromyricetin as the main antioxidant.Conclusion:The developed online extraction HPLC-ABTS method is rapid,stable and green,which provides basis for improvement of the chemical research and product development of Hoveniae Semen.
