GATA3基因rs3824662位点多态性与妊娠期糖尿病相关性研究

Assessment of the association between the rs3824662 of GATA binding protein 3(GATA3)gene and GDM risk

目的探讨GATA3(GATA binding protein 3)基因rs3824662位点单核苷酸多态性与妊娠期糖尿病(gestational diabetes mellitus,GDM)孕妇遗传易感性的相关性。方法选取2012年7月—2013年5月在深圳市布吉人民医院和深圳市龙岗人民医院就诊的GDM孕妇241例为患者组,同期入院的健康孕妇139例为对照组。采用飞行时间质谱技术检测GATA3基因rs3824662位点的基因型,采用二元逻辑回归在3种不同遗传模型中分析该位点与GDM易感性的关系。结果对妊娠年龄和孕前BMI等多项混杂因素进行校正后的加性[OR(95%CI)=1.127(0.490,2.591);P=0.779]、显性[OR(95%CI)=0.976(0.638,1.492);P=0.910]及隐性[OR(95%CI)=1.153(0.515,2.584);P=0.729]遗传模型均显示rs3824662位点多态性与GDM无相关性。结论GATA3基因rs3824662位点单核苷酸多态性(SNP)与GDM的发生在中国人群中可能不具有相关性。

Objective This study aimed to investigate the association between the single nucleotide polymorphism site of rs3824662 in GATA3 and the genetic susceptibility to gestational diabetes mellitus in Chinese population.Methods A total of 241 cases of GDMpregnant women and 139 healthy pregnant women were recruited from July 2012 to May 2013 in Shenzhen Longgang People's Hospital and Shenzhen Buji People's Hospital.The genotypes of rs3824662 were examined by time-of-flight mass spectrometry and the association between rs3824662 and GDM susceptibility was assessed by binary logistic regression in three different genetic models.Results The rs3824662 in GATA3 gene was not associated with GDMsusceptibility in additive[OR(95%CI)=1.127(0.490,2.591);P=0.779],dominant[OR(95%CI)=0.976(0.638,1.492);P=0.910]and recessive[OR(95%CI)=1.153(0.515,2.584);P=0.729]models after adjusting confounding factors including gestational age and pre-pregnancy BMI.Conclusion We observed no association between the single nucleotide polymorphism site of rs3824662 in GATA3 gene and GDM susceptibility in Chinese pregnant women.
