一种同时鉴定多种甜菊糖苷的薄层色谱分析方法 被引量:6

摘要 目的:在工业大规模的生产中,实现对甜菊糖苷产品简易和高效检测;方法:分为单向展开与二维垂直展开两种方法。均以GF254硅板为固定相,展开剂分别是展1-[氯仿∶甲醇∶水=65∶35∶10下层液,1mL甲酸],展2-[正丁醇∶醋酸∶水=4∶1∶5上层液]。结果:试验结果表明所建立的单维薄层色谱方法能快速地实现四种甜菊糖苷的有效分离,色谱斑点清晰。且在此基础上,二维薄层色谱法可用于收集四种甜菊糖苷单体。结论:所建立检测方法具备显著的优势,通过方法学考察,其专一性、重复性、杂质耐受性和实用性都非常理想,适用于工业生产流程中的甜菊糖苷产品检测。 Objective:To apply a simple and efficient detection of steviol glycosides in large-scale industrial production.Methods:It can be divided into one-way expansion and two-dimensional vertical expansion.GF254 silicon plate was used as the stationary phase,and the developing agents were 1-[chloroform∶methanol∶water=65∶35∶10 lower liquid,1mL formic acid]and developing solvent 2-[n-butanol∶acetic acid∶water=4∶1∶5 upper liquid].Results:The results showed that the established method could quickly separate four steviol glycosides and chromatography spots were clear.Based on the previous test,two-dimensional thin layer chromatography was used to collect four stevioside components.Conclusion:The advantage of the established detection method was obvious,its specificity,repeatability,impurity tolerance and usefulness are all highly desirable and suitable for testing of stevioside products in industrial manufacturing.
