沉默UBA2对三阴性乳腺癌细胞系MDA-MB-231增殖和迁移的影响

摘要 目的探讨UBA2在乳腺癌预后中的价值及其对三阴性乳腺癌细胞系MDA-MB-231增殖、迁移和凋亡的影响。方法利用Kaplan-Meier Plotter数据库和人类蛋白质图谱数据库,研究UBA2在乳腺癌中的表达情况和预后价值。通过沉默UBA2,探究其对MDA-MB-231细胞增殖、迁移和凋亡的影响。结果生物信息学分析结果显示,乳腺癌组织中UBA2表达高于正常乳腺组织,且UBA2高表达的患者其预后较UBA2低表达的患者差。通过在MDA-MB-231中沉默UBA2发现,敲低UBA2后细胞增殖和迁移受到抑制,但对凋亡无影响。结论UBA2在乳腺癌中高表达,与预后不良有关。沉默UBA2可以抑制三阴性乳腺癌细胞的增殖和迁移。 Objective To explore the prognostic value of UBA2 in breast cancer and the role of this gene in the proliferation,migration,and apoptosis of the triple-negative breast cancer cell line MDA-MB-231.Methods The Kaplan-Meier Plotter and Human Protein Atlas databases were used to evaluate the expression and prognostic value of UBA2 in breast cancer.UBA2 was silenced to determine its role in the proliferation,migration,and apoptosis of MDA-MB-231 cells.Results Bioinformatic analysis showed that the expression level of UBA2 was higher in the breast cancer tissues than in the normal tissues,and the patients with high UBA2 expression had worse prognosis than those with low UBA2 expression.Silencing the UBA2 expression in MDA-MB-231 cells inhibited their proliferation and migration,but had no effect on their apoptosis.Conclusion UBA2 is highly expressed in breast cancer tissues and associated with poor prognosis.Silencing UBA2 inhibits the proliferation and migration of triple-negative breast cancer cells.
