mtDNA拷贝数在黑色素瘤术后复发患者中的表达及与临床病理指标的关系

Expression of mtDNA Copy Number in Patients with Recurrence after Surgery for Melanoma and Its Relationship with Clinicopathological Indicators

目的探讨线粒体DNA(mtDNA)拷贝数在黑色素瘤术后复发患者中的表达及与临床病理指标的关系。方法选取2018年5月—2020年8月收治的黑色素瘤术后复发患者40例为病例Ⅰ组及黑色素瘤初发患者48例为病例Ⅱ组;另选取同期健康受试者50例为对照组。用实时定量聚合酶链式反应测定病例Ⅰ、Ⅱ组外周血、肿瘤组织、癌旁组织和对照组外周血中mtDNA拷贝数。分析mtDNA拷贝数在不同TNM分期患者中的表达及与术后复发黑色素瘤患者临床病理指标的关系。结果病例Ⅰ组及Ⅱ组肿瘤组织mtDNA拷贝数均高于癌旁组织,且病例Ⅰ组肿瘤组织mtDNA拷贝数高于Ⅱ组,差异有统计学意义(P<0.05)。病例Ⅰ组及Ⅱ组外周血mtDNA拷贝数高于对照组,且病例Ⅰ组高于病例Ⅱ组,差异有统计学意义(P<0.05)。病例Ⅰ组不同TNM分期肿瘤组织mtDNA拷贝数均高于病例Ⅱ组(P<0.05)。肿瘤组织mtDNA拷贝数与术后复发黑色素瘤患者淋巴结转移密切相关(P<0.05)。结论mtDNA拷贝数与黑色素瘤患者的发生、复发及转移具有密切关系。

Objective To explore the expression of mitochondrial DNA(mtDNA)copy number in patients with recurrence after surgery for melanoma and its relationship with clinicopathological indicators.Methods Patients with melanoma recurrence after surgery from May 2018 to August 2020 were selected as caseⅠgroup(n=40)and patients with new-onset melanoma as caseⅡgroup(n=48).Another 50 healthy subjects during the same period were selected as the control group.The mtDNA copy number was determined by real-time quantitative PCR.The expression of mtDNA copy number in patients with different TNM stages and its relationship with clinicopathological indicators of melanoma patients with postoperative recurrence was analyzed.Results The copy numbers of mtDNA in tumor tissues of caseⅠgroup andⅡgroup were higher than those of adjacent tissues,and the copy numbers of mtDNA in tumor tissues of caseⅠgroup were significantly higher than those of caseⅡgroup(P<0.05).The mtDNA copy numbers in peripheral blood of the caseⅠgroup andⅡgroup were higher than those of the control group,which were significantly higher in the caseⅠgroup than in the caseⅡgroup(P<0.05).The copy numbers of mtDNA in tumor tissues of different TNM stages in caseⅠgroup were higher than those in caseⅡgroup(P<0.05).The mtDNA copy numbers of tumor tissue were closely related to lymph node metastasis in patients with recurring melanoma after surgery(P<0.05).Conclusion mtDNA copy number is closely related to the occurrence,recurrence and metastasis of melanoma.
