原核表达家蝇Phormicin蛋白对金黄色葡萄球菌的抑菌活性分析 被引量:2

摘要 家蝇Phormicin是一类具有广普抗菌活性的抗菌肽,属于防御素家族。本研究采用原核表达法纯化并获得家蝇Phormicin A、Phormicin B蛋白,将其作用于金黄色葡萄球菌标准株,通过微量液体稀释法测定其最小抑菌浓度分别为0.625 mmol/L和0.469 mmol/L。进一步绘制了二者对金黄色葡萄球菌的时间杀菌曲线,并利用牛津杯法进一步验证了二者的抗菌效果。结果显示,二者对金黄色葡萄球菌都具有一定的抑菌效果。本研究为进一步探索Phormicin在家蝇天然免疫和防御中的机制提供理论参考。 Phormicin in Musca domestica is a kind of antimicrobial peptide with broad antibacterial activity,belonging to the defense in family.In this study,Phormicin A and B proteins of housefly were purified by prokaryotic expression method.It was applied to the standard strain of Staphylococcus aureus,and its minimum inhibitory concentration(MIC)was determined to be 0.625 mmol/L and 0.469 mmol/L by trace liquid dilution method.The time growth curve of S.aureus was further drawn.The Oxford cup method was used to verify their antibacterial effect further.These results showed that both of them had certain antibacterial effects on S.aureus.This study provides a theoretical reference for further exploring the mechanism of Phormicin in innate immunity and defense process of housefly.
