摘要
采用乙醇回流法提取香椿树皮总黄酮,通过L_(9)(3^(4))正交试验优化总黄酮的提取工艺,并就总黄酮对1,1-苯基-2-苦基肼自由基(DPPH·)和羟基自由基(·OH)的清除作用进行初步研究。结果表明:总黄酮最佳提取工艺件为乙醇体积分数40%、料液比1∶20 (g/mL)、提取温度80℃、提取时间90 min,此时香椿树皮总黄酮平均提取率为15.30%;总黄酮对DPPH·和·OH的清除率可分别达到74.76%和72.53%,说明香椿树皮总黄酮具有一定的抗氧化活性,且其质量浓度与抗氧化性具有量效关系。
Total flavonoids in bark of Toona sinensis were extracted using an ethanol refluxing method and the extraction procedure was optimized with L_(9)(3^(4)) orthogonal experiments. The scavenging effects of total flavones on 1,1-phenyl-2-picrylhydrazine free radicals (DPPH·) and hydroxyl free radicals(· OH)were preliminarily studied. The results showed the optimum extraction conditions was following: the volume fraction of ethanol as extractant was 40 %,the ratio of solid to liquid was 1∶20 (g/m L), the extraction temperature was 80 ℃ and the extraction time was 90 min. Under these conditions, the average extraction rate of total flavonoids in bark of Toona sinensis reached 15. 30 %. The scavenging effects of total flavonoids on DPPH· and· OH were 74. 76 % and 72. 53 % respectively. The results showed that the total flavonoids in bark of Toona sinensis had certain antioxidant activity, and the concentration and antioxidant effect has the dose-effect relationship.
