长链非编码RNA小核仁RNA宿主基因3在肺腺癌组织中的表达及其临床意义

Expression of long-chain noncoding RNA small nucleolar RNA host gene 3 in lung adenocarcinoma tissue and its clinical significance

摘要 目的探讨肺腺癌组织中长链非编码RNA小核仁RNA宿主基因3(lncRNA SNHG3)的表达及临床意义。方法收集120例肺腺癌患者术后癌组织及癌旁组织作为样本,检测样本中SNHG3的表达并分析其与患者临床特征的相关性,利用RNA干扰技术对SNHG3的表达进行干扰,通过噻唑蓝(MTT)实验及Transwell实验记录肺腺癌细胞的生长及侵袭情况。结果肺腺癌组织样本中lncRNA SNHG3的相对表达量(126.83±13.49)明显高于癌旁组织(69.15±9.43),差异具有统计学意义(t=38.389,P<0.001)。发生淋巴结转移、临床分期高、肿瘤直径≥5 cm的肺腺癌组织中lncRNA SNHG3的相对表达量越高(P<0.05)。转染siRNA后SNHG3的表达量明显低于未干扰组(P<0.05)。MTT实验结果显示:与对照组比较,肺腺癌GLC-82细胞增殖能力明显降低(P<0.05)。Transwell实验结果显示,与对照组比较,肺腺癌GLC-82细胞侵袭能力明显降低(P<0.05)。结论肺腺癌组织中lncRNA SNHG3呈高表达,其可能参与了肺腺癌的发生进展。 Objective To investigate the expression and clinical significance of long non-coding RNA small nucleolar RNA host gene 3(lncRNA SNHG3)in lung adenocarcinoma tissues.Methods Postoperative cancer tissues and adjacent tissues were collected from 120 patients with lung adenocarcinoma as samples.SNHG3 expression was detected in samples and its correlation with clinical characteristics of patients was analyzed.RNA interference technology was used to interfere with SNHG3 expression,and the growth and invasion of lung adenocarcinoma cells were recorded by thiazole blue(MTT)and Transwell experiments.Results The relative expression of lncRNA SNHG3 in lung adenocarcinoma tissue samples(126.83±13.49)was significantly higher than that in adjacent tissues(69.15±9.43),and the difference was statistically significant(t=38.389,P<0.001).The relative expression of lncRNA SNHG3 was higher in lung adenocarcinoma tissues with lymph node metastasis,high clinical stage,and tumor diameter≥5 cm(P<0.05).The expression of SNHG3 after transfection of siRNA was significantly lower than that of the non-interfering group(P<0.05).The results of MTT experiments showed that compared with the control group,the proliferation ability of lung adenocarcinoma GLC-82 cells was significantly reduced(P<0.05).Transwell results showed that compared with the control group,the invasion ability of lung adenocarcinoma GLC-82 cells was significantly reduced(P<0.05).Conclusion LncRNA SNHG3 is highly expressed in lung adenocarcinoma tissues,which may be involved in the development of lung adenocarcinoma.
