基于高通量测序技术分析感染根结线虫马铃薯根际土壤细菌群落结构 被引量:2

Analysis Bacterial Community of Potato Rhizosphere Soil Caused by Root-Knot Nematodes using High-Throughput Sequencing

摘要 马铃薯根结线虫病害具有侵染性、隐蔽性、突发性和流行性,一旦被感染,将造成严重损失。为进一步分析健康马铃薯与感染根结线虫马铃薯根际土壤细菌的群落结构及多样性,本研究采用高通量测序技术对根际土壤细菌16S rRNA V3-V4区序列进行测序,应用生物信息学手段分析测序结果。结果表明,感染根结线虫的马铃薯根际土壤细菌类群Shannon指数和Chao1指数均比健康植株的低,Simpson指数比健康植株高,说明感病植株根际土壤中细菌群落丰富度和多样性均比未感病植株低。从门(Phylum)科(Family)属(Genus)上分析菌群丰度,可看出感病植株中主要的优势菌门为放线菌门(Actinobacteria);优势菌科为链霉菌科(Streptomycetaceae)和微球菌科(Micrococcaceae);主要的优势菌属为链霉菌属(Streptomyces)和节细菌属(Arthrobacter)。本研究结果为进一步研究马铃薯根际微环境菌群的多样性和平衡性,提高马铃薯产量和质量提供理论基础。 Potato root-knot nematodes(Meloidogyne spp.)is an infectious,concealment,explosive and epidemic disease.It is one of the major obstacles facing the production of potatoesand cause considerable damage.To determine the species composition and community structer of potato rhizosphere soil caused by root-knot nematodes and healthy potato rhizosphere soil,this study used the Illumine Miseq platform by high-throughput sequencing the V3-V4 region of 16S rRNA gene.The results showed that the Shannon index and Chao1 index of rhizosphere soil bacterial groups of potato infected with root-knot nematodes were lower than those of healthy plants,and the Simpson index was higher than that of healthy plants,indicating that the bacterial community richness and diversity were lower than that of uninfected plants.Respectively in the Genus,Family and Phylum,Actinobacteria was the predominant Phylum,Streptomycetaceae and Micrococcaceae was the predominant Family,Streptomyces and Arthrobacter was the predom inant Genus in potato rhizosphere soil caused by root-knot nematodes.These results provide reference for providing yield and quality of potato by increasing the diversity and balance of microflora in potato rhizosphere.
