转录调控因子GntR10在布鲁氏菌感染细胞和小鼠模型中的毒力作用研究

摘要 探讨转录调控因子GntR10在布鲁氏菌感染过程中的毒力作用.采用分子生物学方法,构建牛种布鲁氏菌2308 gntR10基因突变株(2308ΔGntR10)和互补株(2308ΔGntR10-C),利用突变株和互补株感染上皮细胞系(HPT-8)、小鼠巨噬细胞系(RAW 264.7)和小鼠模型(BALB/c),检测突变株在细胞和小鼠模型中的胞内存活能力和毒力.2308ΔgntR10突变株在细胞模型中的胞内存活能力显著降低,在小鼠模型中是减毒的.转录调控因子GntR10的缺失影响了布鲁氏菌在细胞和小鼠内的毒力,并且在布鲁氏菌感染期间,GntR10基因与细菌的存活和胞内繁殖有关. To explore the virulence of transcriptional regulator GntR10 in Brucella infection.2308ΔgntR10 mutant and 2308ΔgntR10-C complementary strain were constructed by molecular biology.The epithelial cell lines(HPT-8),murine macrophage cell lines(RAW 264.7)and mice models(BALB/c)were infected by 2308ΔgntR10 and 2308ΔgntR10-C.The intracellular ability and virulence of mutant in cell and mice models were detected.The intracellular survival of 2308ΔgntR10 mutant was significantly reduced in cells.2308ΔgntR10 was attenuated in mice models.Mutation of gntR10 affects the virulence of B.abortus to cells and mice.The gntR10 gene is essential for survival and intracellular multiplication of B.abortus during its infection.
