摘要
目的探讨β-谷甾醇(β-sitosterol)对高糖诱导的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)损伤的作用及其机制。方法将HUVECs分为对照组、单纯高糖组、高糖+10μmol/Lβ-谷甾醇组和高糖+50μmol/Lβ-谷甾醇组,用中性红摄取法检测细胞活力,用2’,7’-二氯荧光二乙酸酯(DCF-DA)测定法检测细胞内ROS水平,用二氨基荧光素-FM二乙酸(DAF-FM DA)标记法检测细胞内NO水平,用Western blot法检测细胞的NF-κB p65、iNOS和COX-2水平。结果在高糖条件下,β-谷甾醇可升高HUVECs细胞活力,并降低细胞内ROS生成和NO水平,并可下调NF-κB p65、iNOS和COX-2水平。结论β-谷甾醇可通过降低氧化应激和NF-κB活性以及iNOS和COX-2水平来减轻高糖诱导的HUVECs细胞损伤。
Objective To investigate the effect ofβ-sitosterol on high glucose-induced injury in human umbilical vein endothelial cells(HUVECs)and its mechanism.Methods HUVECs were divided into the control group,the high-glucose alone group,the high-glucose+10μmol/Lβ-sitosterol group and the high-glucose+50μmol/Lβ-sitosterol group.Cell viabilities were measured by neutral red uptake assay;intracellular ROS production were detected by 2’,7’-dichlorodihydrofluorescein diacetate(DCF-DA)assay;intracellular NO levels were detected by diaminofluorescein-FM diacetate(DAF-FM DA)labelling method;the levels of NF-κB p65,iNOS and COX-2 were detected by Western blot.Resultsβ-sitosterol increased the cell viability of HUVECs,decreased the ROS and NO production,and supressed the expression of NF-κB p65,iNOS and COX-2 in them under high glucose condition.Conclusionβ-sitosterol alleviates high glucose-induced injury by reducing oxidative stress,NF-κB activity and the expression of iNOS and COX-2 in HUVECs.
