摘要
目的探讨桑辛素在胶质瘤细胞的抗癌作用。方法用CCK-8检测细胞增殖情况,利用Traswell试验进行细胞迁移分析,用异硫氰酸荧光素(Annexin V-FITC)凋亡检测试剂盒检测细胞凋亡情况,采用免疫印迹试验(Western blotting)和实时荧光定量PCR(RT-qPCR)检测靶基因mRNA和蛋白水平的变化。结果桑辛素能明显抑制U251细胞的增殖活性(P<0.05),其中100和1000μg/mL桑辛素的抑制效果最佳。100μg/mL的桑辛素能明显抑制U251细胞的迁移能力(P<0.05),并诱导细胞凋亡(P<0.05)。Wnt/β-catenin信号通路参与桑辛素诱导的抗肿瘤活性,桑辛素可下调Wnt/β-catenin信号通路相关蛋白(β-catenin和Cyclin D1)的表达(P<0.05)。结论桑辛素可通过Wnt/β-catenin信号途径抑制胶质瘤细胞的生长,因此,桑辛素可能是一种潜在的胶质瘤抗癌药物。
Objective To investigate the anticancer effect of moristin on glioma cells.Methods Cells proliferation was examined by CCK-8.Migration assays were performed using transwell assay.Apoptotic cells were detected using the Annexin V-FITC apoptosis detection kit.Western blotting and real time fluorescence quantitative PCR(RT-qPCR)were used to exam the change of target gene in m RNA and protein level.Results Morusin can obviously inhibited the proliferation activity of U251 cells(P<0.05),of which 100 and 1000μg/mL had the best inhibitory effect.100μg/mL of morusin can obviously inhibited the migration ability of U251 cells and induce cell apoptosis(both P<0.05).Wnt/β-catenin signaling pathway was involved in anti-tumor activity induced by moristin,and morusin can down-regulate the expression of Wnt/β-catenin signaling pathway-related proteins(β-catenin and Cyclin D1)(P<0.05).Conclusion Morusin can inhibit the growth of glioma cells through Wnt/β-catenin signal pathway.Thus,morusin could be a potential anti-cancer agent for glioma.
