摘要
紫甘蓝色素,具有广泛的开发应用前景。而色素对酸、碱及化学物质的稳定性直接关系到色素的应用范畴。本综合实验采用微波萃取和柱层析法分离提纯紫甘蓝色素,再通过加入不同pH值或不同金属离子的溶液,用紫外可见光谱法测试两者对紫甘蓝色素稳定性的影响。让学生了解紫甘蓝提取工艺和提纯方法,明白微波萃取法和色谱法分离提纯有机物的原理,掌握微波萃取和柱层析操作技术,学会分析不同酸度、金属离子对紫甘蓝天然色素的影响。
The cabbage red pigment extracted from purple cabbage has a wide application prospect.The stability of pigment for acid,alkali and chemical substances is directly related to the pigment application.In this comprehensive experiment,anthocyanin is separated from purple cabbage pigment by the microwave extraction and then purified by column chromatography.The influence of pH values and metal ions on the pigment stability is investigated by UV-visible absorption spectrometer.This experiment enables students to understand extraction technology and purification method of purple cabbage pigment,understand the principle of microwave extraction and chromatography separation and purification,master the microwave extraction and column chromatography operation technology,learn to analyze the effect of different acidity and metal ions on purple cabbage pigment.
