痹祺胶囊治疗胶原诱导型关节炎大鼠的作用机制研究

Study on mechanism of Biqi Capsule in treating collagen induced arthritis rats

目的探究痹祺胶囊对胶原诱导型关节炎(collagen induced arthritis,CIA)大鼠中软骨寡聚基质蛋白(cartilage oligomeric matrix protein,COMP)表达的影响。方法随机选取10只SD大鼠作为对照组,其余大鼠建立CIA模型,CIA大鼠随机分为模型组,痹祺胶囊中、高剂量(0.6、0.9g/kg)组,甲氨蝶呤(1.67mg/kg)组,连续给药4周,观察大鼠的一般情况和足肿胀变化,检测大鼠血清中COMP水平、滑膜中COMP蛋白表达、软骨中COMP mRNA表达。结果与对照组比较,模型组大鼠血清中COMP水平、滑膜中COMP蛋白表达、软骨中COMP mRNA表达均显著升高(P<0.001);与模型组比较,各给药组大鼠血清中COMP水平、滑膜中COMP蛋白表达、软骨中COMP mRNA表达均显著降低(P<0.001)。结论痹祺胶囊可能通过下调COMP表达,抑制炎性细胞浸润、滑膜增生和关节软骨破坏,从而治疗类风湿关节炎。

Objective To investigate the effect of Biqi Capsule(痹祺胶囊) on the expression of cartilage oligomeric matrix protein(COMP) in CIA rats. Methods A total of 10 rats were randomly selected as control group, the other rats were used to establish CIA model, CIA rats were randomly divided into model group, low-, medium-dose Biqi Capsule(0.6, 0.9 g/kg) groups and methotrexate(1.67 mg/kg) group. After administration for 4 weeks, the changes of paw swelling in rat were observed;The level of COMP in serum, expression of COMP in synovium and mRNA level of COMP in cartilage were tested. Results Compared with control group, level of COMP in serum, expression of COMP in synovium and m RNA level of COMP in cartilage were significantly increased in model group(P < 0.001);Compared with model group, level of COMP in serum, expression of COMP in synovium and m RNA level of COMP in cartilage were decreased in treatment groups(P < 0.001). Conclusion Biqi Capsule may prevent inflammatory infiltration, synoviocyte proliferation, cartilage destruction by inhibiting the expression of COMP, and then treat rheumatoid arthritis.
