小麦BES1转录因子全基因组鉴定与分析 被引量:2

摘要 为了解小麦中BES1基因家族功能,根据已公布的小麦基因组信息(IWGSC v1.1),对小麦BES1基因家族进行全基因组鉴定。根据已报道的BES1基因,采用同源比对法检索小麦基因组中的BES1家族基因,pfam鉴定并进行编号,通过ExPASy Proteomics Server预测小麦BES1氨基酸序列的基本信息,利用Cell-PLoc进行亚细胞定位预测,采用MEGA 7软件构建进化树,利用R软件包pheatmap绘制启动子的热图和circlize绘制同源关系图谱。结果表明,小麦共有15个BES1基因,被分为4组;小麦BES1编码的蛋白质等电点为8.13~9.4,不稳定指数为50.78~69.99;小麦BES1启动子区域一共含有838个顺式元件,471(56.2%)个与生长发育有关,201(24%)个与非生物/生物胁迫有关,166(19.8%)个与激素反应有关;与普通小麦相关的同源物共52对,有13对(25%)旁系同源物和39对(75%)直系同源物。表明小麦BES1基因家族包括15个成员,全部为碱性蛋白质和不稳定蛋白质,小部分(13对25%)来源于自身的进化,大部分(39对75%)来源于3种亚基因组供体小麦。 In order to understand the function of the BES1 gene family in wheat,the wheat BES1 gene family was identified according to the published wheat genome information(IWGSC v1.1).Based on the reported BES1,the homology comparison method was used to search the BES1 family genes in the wheat genome,pfam identification and it was numbered finally,the basic information of wheat BES1 protein amino acid sequence was predicted by ExPASy Proteomics Server,Cell-PLoc was used to predict subcellular location,MEGA7 software was used to build evolutionary tree,the heat map of the promoter was drawn by the R software package,the homology map was drawn by using the R software package.The results showed that there were 15 BES1 genes in wheat,which were divided into 4 groups;the isoelectric point of the protein encoded by wheat BES1 was 8.13-9.4,and the instability index was 50.78-69.99;The wheat BES1 promoter region contained a total of 838 cis-elements,471(56.2%)was related to growth and development,201(24%)was related to abiotic/biological stress,and 166(19.8%)was related to hormone response;there were 52 pairs of homologues related to common wheat,including 13 pairs(25%)of paralogues and 39 pairs(75%)of orthologous.The results show that the wheat BES1 gene family includes 15 members,all of which are basic and unstable proteins,a small part(13 vs.25%)is derived from its own evolution,andmost(39 vs.75%)of them are derived from three sub-genome donors species.
