摘要
目的:通过网络药理学方法探究丹参水溶性成分治疗冠心病的作用机制。方法:使用中国知网(CNKI)、Pubchem、化源网、化学专业数据库,搜集丹参中活性较高的水溶性成分,并通过SwissTargetPrediction数据库预测成分的作用靶点;利用Genecards、DisgeNET 2个数据库获取疾病靶点;利用String数据库构建PPI网络,根据Degree值筛选核心靶点;采用Metascape数据库对核心靶点进行GO生物功能和KEGG信号通路富集分析,探讨其作用机制。结果:纳入活性成分12个,预测靶点175个,疾病靶点1 873个,PPI筛选核心靶点43个;GO生物功能分析发现,得到3 230个生物过程条目,涉及白细胞迁移、积极调控细胞迁移、对受伤的反应、对氧化应激的反应等;327个细胞组成条目,涉及膜筏、膜面、细胞外基质等;406个分子功能条目,涉及内肽酶活性、激酶结合、蛋白质结构域特异性结合等;KEGG信号通路分析发现,内分泌抵抗、流体剪切应力与动脉粥样硬化、肌动蛋白细胞骨架的调节、肾素-血管紧张素系统等通路参与调控。结论:丹参水溶性成分可促进血液流变,对治疗冠心病具有潜在的积极作用。
Objective:To explore the mechanism of treatment of coronary heart disease with watersoluble ingredients of Salvia miltiorrhiza via network pharmacology.Methods:Use China Knowledge Network(CNKI),Pubchem,Huayuan.com,and chemistry professional databases to collect highly active water-soluble ingredients in Salvia miltiorrhiza,and predict the target of the ingredients through the Swiss Target Prediction database;use two databases,Genecards and Disge NET Disease targets;use the String database to construct a PPI network,and select core targets based on the Degree value;use Metascape database to conduct GO biological function and KEGG signal pathway enrichment analysis of core targets to explore its mechanism of action.Results:This study included 12 active ingredients,175 predicted targets,1873 disease targets,and 43 core targets screened by PPI;GO biological function analysis found that 3230 biological process items were obtained,involving leukocyte migration and active regulation of cells Migration,response to injury,response to oxidative stress,etc.;327 cell composition items,involving processes such as membrane rafts,membrane surfaces,and extracellular matrix;406 molecular function items,involving endopeptidase activity,kinase binding,Protein domain specific binding and other processes;KEGG signaling pathway analysis found that endocrine resistance,Fluid shear stress and atherosclerosis,regulation of actin cytoskeleton,renin-angiotensin system and other pathways participate in regulation.Conclusion:The water-soluble ingredients of Salvia miltiorrhiza can promote blood rheology and have potential positive effects on the treatment of coronary heart disease.
