摘要
目的:应用体素内不相干运动扩散加权成像(IVIM-DWI)技术对沉默miRNA-221抑制裸鼠舌鳞癌移植瘤的早期改变进行监测,探讨miRNA-221表达异常对肿瘤早期的病理生理改变与IVIM参数的相关性.方法:建立人舌鳞癌UM1细胞株裸鼠皮下移植瘤模型12只,实验组和对照组各6只.实验组注射慢病毒转染的稳定沉默miR-221的UM1细胞,对照组注射慢病毒空转染的UM1细胞.细胞接种7,21 d后,进行IVIM-DWI检测,动态测量纯水分子扩散系数(D值)、灌注相关扩散系数(D值)及灌注分数(f值).于第21天时间点处死全部裸鼠并获取肿瘤标本,将所取肿瘤标本进行常规HE和CD34免疫组化染色.结果:裸鼠皮下接种沉默miR-221表达的UM1细胞后,在第7天和21天,实验组的D值均显著高于对照组,差异具有统计学意义(P<0.01);实验组的D值、f值均显著低于对照组,差异具有统计学意义(P<0.05).结论:IVIM-DWI相关参数中的D值、D值和f值,可以对沉默miR-221抑制裸鼠舌鳞癌移植瘤的早期疗效进行监测与定量评估;IVIM的相关参数D值、D值和f值变化与病理改变和CD34关系密切.
Objective:To monitor the treatment of early changes in tongue subcutaneous transplanted tumor in nude mice by silencing miRNA-221,using the IVIM-DWI technique and to investigate the correlation between the early pathological and physiological changes in tumor and the IVIM parameters after silencing miRNA-221 treatment.Methods:Twelve nude mice were randomly divided into examination group and control group.The experimental group mice were subcutaneously injected with lentiviral transfected UM1 cells which were stably low expression with miR-221,and the control group mice were subcutaneously injected with lentiviral empty transfected UM1 cells.All of the nude mice were received the intravoxel incoherent motion diffusion weighted imaging(IVIM-DWI)at 7 d and 21 d.The pure water molecular diffusion coefficient(D),perfusion-related diffusion coefficient(D value)and perfusion fraction(f value)values were measured dynamically.And all of them were sacrificed at 21 d,and the tumor samples were treated with the routine HE and CD31 immunohistochemical staining.Results:After subcutaneous inoculation of UM1 cells by silencing miR-221 in nude mice,the D value of the experimental group was significantly higher than that of the control group on 7 d and 21 d,with statistically significant difference(P<0.01).While the D and f values of the experimental group were both significantly lower than that of the control group on 7 d and 21 d,with statistically significant difference(P<0.05).Conclusion:Among the IVIM-DWI parameters,D,D and f can dynamically monitor and quantitatively evaluate the early effect of silencing miR-221 on tongue subcutaneous transplanted tumor in nude mice.And the changes in D,D and f values are closely related to the pathology and CD34.
