miR-203和survivin在尖锐湿疣患者组织中的表达及其与HPV分型的相关性

Expressions of miR-203 and survivin in condyloma acuminatum and their correlations with HPV typing

目的探讨微小RNA-203(miR-203)和survivin在尖锐湿疣(CA)患者组织中的表达及其与人乳头瘤病毒(HPV)分型的相关性。方法选取2015年6月至2018年6月扬州大学附属医院收治的100例CA患者作为研究对象。将这100例CA患者设为CA组,另选取50例在扬州大学附属医院行包皮环切术患者的正常包皮组织标本作为对照组。采用实时荧光定量PCR检测组织中miR-203、survivin mRNA表达,采用免疫组化法检测组织中survivin蛋白表达情况,观察不同分组患者miR-203、survivin表达情况及二者表达相关性。结果100例CA患者中HPV阳性90例(90.00%),其中高危型31例(34.44%),低危型59例(65.56%);高危型组miR-203 mRNA表达水平明显低于低危型组、对照组,低危型组低于对照组(P<0.05);高危型组survivin mRNA表达高于低危型组、对照组,低危型组高于对照组(P<0.05);高危型组survivin阳性率为70.97%,高于低危型组(40.68%)、对照组(6.00%)(P<0.05);CA患者组织中miR-203与survivin表达呈负相关(P<0.05)。结论CA患者组织中miR-203低表达,survivin高表达,二者表达呈负相关性,并与HPV分型相关。

Objective To investigate the expression of microRNA-203(miR-203)and survivin in condyloma acuminatum(CA)and its correlation with human papillomavirus(HPV)typing.Methods 100 patients with CA admitted to the Affiliated Hospital of Yangzhou University from June 2015 to June 2018 were selected as the CA group,and another 50 normal prepuce specimens undergoing circumcision as the control group.the expressions of miR-203 and survivin mRNA was detected by real-time fluorescence quantitative PCR,the expression of survivin was detected by immunohistochemistry,and the expressions of miR-203 and survivin in different groups and their correlation were observed.Results Among the 100 CA patients,90 cases(90.00%)were HPV positive,including 31 cases(34.44%)of high-risk type and 59 cases(65.56%)of low-risk type.The mRNA expression level of miR-203 in high-risk group was significantly lower than that in low-risk group and control group,and that in low-risk group was lower than that in control group(P<0.05).MRNA expression of survivin in high-risk group was higher than that in low-risk group and control group,and that in low-risk group was higher than that in control group(P<0.05).The survivin positive rate in high-risk group was 70.97%,which was higher than that in low-risk group(40.68%)and control group(6.00%)(P<0.05).There was a negative correlation between miR-203 and survivin expression in the tissues of CA patients(P<0.05).Conclusions There was a low expression of miR-203 and a high expression of survivin in the tissues of patients with CA,and the expressions of the two were negatively correlated and correlated with HPV typing.
