补肾活血汤对多囊卵巢综合征不孕症大鼠卵巢超微结构的影响及助孕效果观察

Effect of Bushenhuoxue Decoction on ovary ultrastructure in infertile rats with polycystic ovary syndrome and its effect on assisting pregnancy

目的探讨补肾活血汤对多囊卵巢综合征(PCOS)不孕症大鼠卵巢超微结构的影响,并观察其助孕效果。方法选取80只雌性SD大鼠随机分为五组,每组16只。除N组外均建立PCOS不孕症模型,五组分别给予无菌生理盐水(N组)、无菌生理盐水(M组),以及1.0g/mL(LD组)、2.0g/mL(MD组)和4.0g/mL(HD组)三种剂量的补肾活血汤灌胃。对比干预后血清性激素水平;观察各组卵巢超微结构及卵巢凋亡颗粒细胞数;将各组剩余大鼠待性成熟后与SD性成熟雄鼠按照2∶1同笼配繁,统计妊娠成功率。结果各组血清性激素水平比较,差异均具有统计学意义(P<0.05),血清孕酮(P)、黄体生成素(LH)比较,M组>LD组>MD组>HD组>N组,血清雌二醇(E 2)比较,N组>HD组>MD组>LD组>M组。干预后N组卵巢镜下黄体和卵泡均发育状态均正常,M组均显著改变,LD、MD和HD组均有所好转,且呈剂量依赖性。五组大鼠干预后卵巢凋亡颗粒细胞数比较,差异具有统计学意义(P<0.05),且M组>LD组>MD组>HD组>N组。五组剩余大鼠妊娠成功率比较,差异具有统计学意义(P<0.05),其中N组>HD组>MD组>LD组>M组。结论补肾活血汤可改善PCOS不孕症大鼠性激素水平和卵巢超微结构,达到良好的助孕效果,且呈剂量依赖。

Objective To explore the effect of Bushenhuoxue Decoction on ovary ultrastructure in infertile rats with polycystic ovary syndrome and its effect on assisting pregnancy.Methods 80 female SD rats were randomly divided into 5 groups with 16 rats in each group.PCOS infertility models were established except for group N.The 5 groups were given sterile normal saline(group N),sterile normal saline(group M),and 1.0g/mL(group LD),2.0g/mL(group MD)and 4.0g/mL(group HD)doses of Bishenhuoue Decoction,respectively.Serum sex hormone levels were compared after intervention.The ultrastructure and the number of apoptotic granulosa cells of ovary in each group were observed.After sexual maturation,the remaining rats in every group were mated with SD male rats in the same cage according to the ratio of 2∶1,and the pregnancy success rates were calculated.Results There were significant differences in the levels of serum sex hormone among the groups(P<0.05).As to the levels of serum progesterone(P)and luteinizing hormone(LH),M group>LD group>MD group>HD group>N group;the levels of serum estradiol(E 2),N group>HD group>MD group>LD group>M group.After intervention,luteal and follicular development were normal in N group,and significantly changed in the M group,and the LD,MD and HD groups were all improved in a dose-dependent manner.There was a significant difference in the number of apoptotic granulosa cells among the groups(P<0.05),and M group>LD group>MD group>HD group>N group.There was a significant difference in pregnancy success rates of the remaining rats among the 5 groups(P<0.05),and the N group>HD group>MD group>LD group>M group.Conclusions Bushenhuoxue Decoction can improve the sex hormone levels and ovarian ultrastructure of rats with PCOS infertility,and achieve a good effect of assisting pregnancy,which is dose-dependent.
