摘要
利用直接测序法研究猪C-X-C基序趋化因子10(C-X-C motif chemokine ligand 10,CXCL10)基因5′侧翼区域多态性,并开展5′侧翼区域和3′UTR遗传变异位点群体遗传结构分析及其与哺乳仔猪腹泻和生长性状关联分析。发现在CXCL10基因5′侧翼区域存在13个SNPs位点,建立针对5′侧翼区域SNP rs337535939 NdeⅠPCRRFLP分型技术。在民猪和长白猪群体中作SNP rs337535939和SNP c.407T>C与表型性状关联分析。结果表明,G等位基因在民猪和长白猪群体中基因频率分别为0.51和0.82;GG型民猪和长白猪35日龄断奶重和日增重显著高于AA型个体。GG型长白猪具有低腹泻指数。SNP c.407T>C对民猪和长白猪腹泻和生长速度无影响。综上,选育GG型个体可降低腹泻发生率,提高仔猪日增重,有望作为新遗传标记。
In this study,direct sequencing was used to study the polymorphism in the 5'flanking region of the porcine C-X-C motif chemokine ligand 10(CXCL10)gene,and to expand the population genetic structure of the 5'flanking region and 3'UTR genetic variation sites analysis and its correlation analysis with diarrhea and growth traits of suckling piglets.It was found that there were 13 SNPs in the 5′flanking region of CXCL10 gene,and a NdeⅠPCR-RFLP technology was established to detect the SNP rs337535939 in the 5′flanking region.The association analysis between SNP rs337535939(and SNP c.407T>c in 3′UTR)and phenotypic traits was carried out in Min and Landrace piglets.The results of genetic structure analysis showed that the gene frequency of G allele was 0.51 in Min piglets and 0.82 in Landrace piglets.The association analysis showed that,for the SNP rs337535939,the average daily gain(ADG)and 35-day body weight of Min and Landrace piglets with GG genotype were significantly higher than that of AA genotype(P<0.05).The Landrace piglets with GG genotype had low diarrhea score.For the SNP c.407 T>C,the difference between phenotype values of different genotypes was not significant in both Min and Landrace piglets.The research showed that GG individuals could be selected to reduce the occurrence of diarrhea and increase the average daily gain of piglets could be used as a new genetic marker.
