基于网络药理学方法的芍药甘草汤治疗神经性疼痛作用机制探讨 被引量:7

摘要 目的基于网络药理学方法探讨芍药甘草汤治疗神经性疼痛的作用机制。方法基于TCMSP、DrugBank、TTD数据库分别建立了芍药甘草汤的化学成分-靶点网络和疾病靶点网络,基于Cytoscape 3.7.0软件建立了芍药甘草汤的活性成分-作用靶点网络并进行网络拓补分析;利用DAVID数据库对核心作用靶点进行基因本体(GO)功能富集分析和基因组百科全书(KEGG)通路富集分析。结果构建了包含46个节点、2070条边的活性成分-作用靶点网络;核心靶点GO功能注释得到24条富集结果,其中生物过程13条,分子功能4条,细胞组成7条;富集得到10条核心的KEGG通路,包含神经组织交互通路、钙信号通路、唾液分泌、胆碱能突触、脂肪细胞脂解调控、肌动蛋白细胞骨架调控、含血清素的神经突触、心肌细胞肾上腺素能信号、cGMP-PKG信号通路、cAMP信号通路等。结论芍药甘草汤可能是通过调节细胞钙离子传导、胆碱突触信号传递、肾上腺素信号传递等通路协同作用于神经性疼痛,分析结果可为芍药甘草汤在神经性疼痛的药理作用研究及临床应用提供一定的理论依据,有待进一步实验验证。 Objective Mechanism of Shaoyao and Gancao Decoction(芍药甘草汤)in the treatment of neuropathic pain was analyzed and predicted based on network pharmacology.Methods Based on TCMSP,DrugBank and TTD database,the chemical component-target network and disease-target network of Shaoyao Gancao Decoction were established respectively.Cytoscape 3.7.0 software was used to visualize the active component-target network of Shaoyao Gancao Decoction and conduct network extension analysis.Gene ontology(GO)functional enrichment analysis and genome encyclopedia(KEGG)pathway enrichment analysis were performed on the core target using DAVID database.Results The active component-target network with 46 nodes and 2070 edges was established.The GO functional annotation of the core target obtained 24 enrichment results,including 13 biological processes,4 molecular functions and 7 cell components.Ten core KEGG pathways were obtained,including neural tissue interaction pathway,calcium signaling pathway,salivary secretion,cholinergic synapse,lipolysis regulation of adipocytes,cytoskeleton regulation of actin,synapses containing serotonin,adrenergic signal of cardiac myocytes,cgmp-pkg signaling pathway,cAMP signaling pathway,etc.Conclusion Shaoyao Gancao Decoction may relieve neuropathic pain by regulating cell calcium ion conduction synaptic signals,choline,signaling pathways and adrenaline synergy in neuropathic pain.The results of the analysis for Shaoyao Gancao Decoction in neuropathic pain pharmacology research and provide certain theoretical basis for clinical application needs further experiment.
