紫檀芪对肺纤维化小鼠的抗氧化作用研究 被引量:6

摘要 目的研究紫檀芪对博莱霉素诱导的肺纤维化小鼠肺组织中的丙二醛(MDA)和总超氧化物歧化酶(T-SOD)的影响。方法按照体重将小鼠随机分为4组:正常组、模型组、正常给药组和模型给药组,每组10只。向小鼠气管内滴注博莱霉素(5 mg·kg^(-1))建立小鼠肺纤维化模型。从造模后第2天开始,正常组和模型组给予生理盐水,正常给药组和模型给药组给予紫檀芪40 mg·kg^(-1),每天相同时间段灌胃,持续4周。收集小鼠肺组织标本,以硫代巴比妥酸(TBA)法测定小鼠肺组织中的MDA和黄嘌呤氧化酶(羟胺法)测定T-SOD含量。结果正常组、模型组、正常给药组和模型给药组的MDA含量分别为(1.30±0.14),(3.89±0.54),(1.52±0.38)和(2.03±0.27)nmol·mg^(-1);这4组的T-SOD含量分别为(27.47±2.59),(15.36±3.00),(27.21±3.79)和(21.87±2.01)U·mg^(-1)。上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);模型给药组或正常给药组与模型组比较,差异均有统计学意义(均P<0.01)。结论紫檀芪能够降低小鼠肺纤维化的肺组织中MDA和升高T-SOD含量,减轻炎症反应和肺纤维化程度。 Objective To study the effect of Pterostilbene on malondialdehyde(MDA)and total superoxide dismutase(T-SOD)in lung tissue of mice with bleomycin-induced pulmonary fibrosis.Methods The mice were randomly divided into 4 groups according to weight:normal group,model group,normal administration group and model administration group,with 10 mice in each group.Bleomycin(5 mg·kg^(-1))was instilled into the trachea of mice to establish a mouse pulmonary fibrosis model.From the second day,mice in normal group and model group were respectively given 0.9%normal saline through tube feeding,mice in normal administration group and model administration group were respectively given pterostilbene 40 mg·kg^(-1)·d-1 through tube feeding,for 4 weeks.The lung tissue samples of mice were collected.The thiobarbituric acid(TBA)method was used to measure MDA and xanthine oxidase(Hydroxylamine method)to determine the T-SOD content.Results MDA content in normal group,model group,normal administration group and model administration group were(1.30±0.14),(3.89±0.54),(1.52±0.38)and(2.03±0.27)nmol·m-1;the contents of T-SOD in the 4 groups were(27.47±2.59),(15.36±3.00),(27.21±3.79)and(21.87±2.01)U·mg^(-1).Comparison between model group and normal group,the difference of the factors were statistically significant(all P<0.01);comparison between model administration group or normal administration group and model group,the difference of the factors were statistically significant(all P<0.01).Conclusion Pterostilbene can reduce MDA and increase T-SOD content in the lung tissues of pulmonary fibrosis in mice,and reduce inflammation and pulmonary fibrosis degree.
