The relationship between oxidative stress induced by NOX and Alzheimer’s disease

摘要 阿尔兹海默病(Alzheimer’s disease,AD)是一种增龄性进行性发展的神经退行性疾病,以记忆水平下降、认知功能衰退,并伴有神经精神症状和行为障碍为主要特点的临床综合特征。尼克酰胺腺嘌呤二核苷酸磷酸氧化酶(NADPH oxidase,NOX)是机体促成活性氧生成的主要酶类之一,由7种亚型(NOX1、NOX2、NOX3、NOX4、NOX5、DUOX1、DUOX2)组成,统称为NOX家族。氧化应激造成的氧化损伤与AD的发生发展密切相关。现对中枢神经系统中主要神经细胞NOX介导氧化应激与AD病理机制、AD病理的药物干预及运动介导氧化应激与AD的防治最新研究进展进行综述,旨在为临床AD的治疗与防控提供与NOX相关的研究思路与干预策略。 Alzheimer’s disease(AD)is a neurodegenerative disease that develops progressively with aging.It is characterized by decline of memory level and cognitive function,accompanied by neuropsychiatric symptoms and behavioral disorders.Nicotinamide adenine dinucleotide phosphate oxidase(NADPH oxidase,NOX)is one of the main enzymes that contribute to the production of reactive oxygen species.It consists of seven subtypes(NOX1,NOX2,NOX3,NOX4,NOX5,DUOX1,DUOX2),which are collectively referred to as the NOX family.Oxidative damage caused by oxidative stress is closely related to the occurrence and development of AD.This article reviewed the latest research progress of NOX-mediated oxidative stress and AD pathological mechanism in the central nervous system,drug intervention and prevention strategies in AD pathology,exercise-mediated oxidative stress and the prevention and treatment of AD,aiming to provide research ideas and intervention strategies related to NOX for the treatment and prevention of clinical AD.
