基因ALDOB的表达下调与肾癌患者的不良预后的相关性分析 被引量:1

Effects of ALDOB on renal cancer and its gene set enrichment analysis

摘要 目的探讨ALDOB和肾癌的临床病理特征的关系,评价ALDOB对癌症患者的预后评价的意义,预测ALDOB影响肾癌进展的方式。方法2017年收集武汉大学人民医院临床样本进行病理指标的回顾性分析,使用生物信息学方法挖掘数据库GSE40435,验证ALDOB在肾癌中的表达情况,同时研究ALDOB对肾癌患者的预后评价,利用GSEA方法分析预测AL-DOB可能影响肾癌进展的信号通路的基因集。结果在肾癌患者中,ALDOB呈低表达,高分化肾癌组的ALDOB表达较低分化肾癌组更低(P<0.05),且ALDOB低表达的肾癌患者预后明显更差(P<0.05),ALD0B低表达样本富集了与细胞周期,DNA复制、甘氨酸生物合成、p53信号通路、泛素介导的蛋白质水解等信号通路相关的基因集。结论ALDOB是肾癌发展过程中的重要基因,ALDOB对患者的预后有着深远的影响,可以作为新的生物标志物。 Objective To investigate the relationship between ALDOB and clinicopathological features of renal cell carcinoma,to evaluate the significance of ALDOB in evaluating the prognosis of cancer patients,and to predict the way in which ALDOB afects the progression of renal cell carcino-ma.Methods Clinical samples were ollcted for retrospective analysis of pathological indicators in.2017.Bioinformatics was used to mine the database GSE40435 to verify the expression of ALDOB in.renal cell carcinoma.Meanwhile,the prognostic evaluation of ALDOB in renal cell carcinoma patients was studied.GSEA was used to analyze and predict the gene sels of signal pathways that ALDOB may affeet the progression of renal cell carcinoma.Results In renal cell carcinoma,ALDOB expression was low in well-differentiated renal cell carcinoma group and low-differentiated renal cell carcinoma group,and the prognosis of renal cell carcinoma patients with low-expression of ALDOB was worse(P<0.05).Low-expression samples of ALDOB enriched gene sels related to cell cycle,DNA replica-tion,glycine biosynthesis,p53 signaling pathway and ubiquitin.mediated protein hydrolysis.Conclu-sions ALDOB is an important gene in the development of renal cell careinoma.ALDOB has a deep impact on the prognosis of patients and can be used as a new biomarker.
