摘要
采用超高效液相色谱-三重四级杆串联质谱(UPLC-MS/MS)技术在电喷雾离子源(ESI)、正离子多反应监测(MRM)模式下建立蓝莓果实中花青苷类物质的定性和定量检测方法。结果表明,11种花青苷类物质的定量限为0.26~1.50μg/kg,在0~100μg/L范围内呈现很好的线性关系,相关系数(r)为0.9950~0.9996,方法检测性良好。采用本方法对3个蓝莓栽培品种的成熟果实进行测定,分别从‘喜来’和‘蓝冠’中鉴定出11种花青苷,从‘蓝丰’中鉴定出10种花青苷,各种花青苷物质含量在3个蓝莓品种中存在明显差异,‘蓝冠’的总花青苷类物质含量明显高于‘蓝丰’和‘喜来’,‘喜来’中最低。本研究结果可为蓝莓优异种质挖掘和蓝莓花青苷产品开发奠定理论基础。
The anthocyanins in blueberry fruits were qualitatively and quantitatively analyzed by ultra-high performance liquid chromatography coupled with triple quadrupole tandem mass spectrometry(UPLC-MS/MS) under electrospray ion source(ESI) and positive ion multiple reaction monitoring(MRM) mode. The results showed that the quantitative limits of the 11 anthocyanins were 0.26~1.50 μg/kg, which showed a good linearly relationship in the range of 0~100 μg/L with the correlation coefficients(r) of 0.9950~0.9996. The mature fruit samples of three blueberry varieties were analyzed by this method. Eleven anthocyanins were identified from ‘Sierra’ and ‘Languan’, and 10 anthocyanins were identified from ‘Bluecrop’. There were obvious differences in the contents of anthocyanins among the three blueberry varieties. The content of total anthocyanins in ‘Languan’ was higher than that in ‘Bluecrop’ and ‘Sierra’, and that in ‘Sierra’ was the lowest. The results of this study could lay a theoretical foundation for the exploitation of excellent blueberry germplasm and the development of blueberry anthocyanin products.
