摘要
目的探讨脂源性细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)对成熟人脂肪细胞miR-506表达的调控影响。方法采用实时荧光定量多聚核苷酸链式反应方法检测人脂肪前体及成熟脂肪细胞中miR-506的表达水平。并以10、30 ng/mL的TNF-α、IL-6干预成熟脂肪细胞0、4、8、24 h,检测各时间点miR-506的表达水平。结果成熟脂肪细胞中miR-506表达量高于前体脂肪细胞,差异有高度统计学意义(P<0.01)。10 ng/mL的TNF-α、IL-6干预成熟人脂肪细胞4、8、24 h miR-506表达量与干预0 h比较,差异无统计学意义(P>0.05)。30 ng/mL的TNF-α、IL-6干预成熟人脂肪细胞24 h miR-506表达量高于干预0 h,差异均有统计学意义(均P<0.05)。结论miR-506的表达受脂源性细胞因子的调控,为miR-506与肥胖相关胰岛素抵抗和炎症反应相关性的研究提供理论依据。
Objective To investigate the regulatory effect of adipokines tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)on the expression of miR-506 in mature human adipocytes.Methods Real-time fluorescence quantitative polynucleotide chain reaction was used to detect the expression level of miR-506 in human precursor adipocytes and mature adipocytes.TNF-αand IL-6 were treated with 10,30 ng/mL for 0,4,8 and 24 h,and the expression level of miR-506 was detected at each time point.Results The expression level of miR-506 in mature adipocytes was higher than that in precursor adipocytes,and the difference was highly statistically significant(P<0.01).The expression of miR-506 in mature human adipocytes at 4,8 and 24 h after treatment with 10 ng/mL TNF-αand IL-6 were not significantly differents from those at 0 h after treatment(P>0.05).The expression level of miR-506 in mature human adipocytes after treatment with 30 ng/mL TNF-αand IL-6 at 24 h were higher than those after treatment at 0 h,and the differences were statistically significant(all P<0.05).Conclusion miR-506 expression is regulated by adipokines,which provides theoretical basis for the study of the correlation between miR-506 and inflammatory responses and obesity-related insulin resistance.
