摘要 博来霉素(Bleomycin,BLM)一种具有抗肿瘤活性的天然产物,临床应用中具有导致肺纤维化损伤的副作用。本文尝试建立小鼠肺间质纤维化模型简单易行的实验方法。选取C57BL/6小鼠,雌雄性各半,随机分组。BLM组一次性注射盐酸博来霉素200 mg(kg·bw),对照组一次性注射相同剂量的生理性盐水。实验第21 d实验组和对照组的小鼠留取肺组织,观察该实验中的小鼠肺部组织的变化,体重变化以及平均生存率。结果:与对照组雌雄小鼠对比,BLM组小鼠肺部病理的表现主要为典型弥漫性炎、成纤维化局部病理性基质增生和局部肺间质沉积等肺部病理变化特征,雌雄性别无明显差异。本次建模具有小鼠死亡率稍高,且病理变化不均一的特点。更加优化的肺纤维化模型及其分子机制的深入研究有助于开发治疗肺纤维化的药物。 Bleomycin was antitumor agent employed clinically for the treatment of several types of cancer.Objective of this study is to explore the optimal methods of bleomycin to induce pulmonary fibrosis in mice.40 C57BL/6 mice were randomly divided into two groups:BLM male group,BLM female group,control male group and control female group.BLM experimental group was injected with bleomycin 200 mg/(kg·bw)once,while the control group was injected with the same amount of normal saline.On the 21st day,all the mice were killed,and the lung tissues of the mice were taken to observe whether the pathological structure of the lung tissue was changed,and the normal weight and the average survival rate of the mice were observed.Compared with the control group mice,the pulmonary pathological changes in BLM group were characterized by typical diffuse tracheal pneumonia,local pathological stromal hyperplasia and fibrocyte of fibroblast,deposition of exogenous stroma in local pulmonary interstitium and local alveolar cavity.Seven BLM model mice died and the mortality rate was 35%.In this method experiment,the C57BL/6 mouse model of early pulmonary fibrosis was established by injecting single-large-dose bleomycin(200 mg/(kg·bw))in tail vein,although the early death rate of mice is little high,and showed inhomogeneous fibrosis in the lung.Then the simple and effective technical to establish fibrosis model and researches of its mechanism will make it helpful to research and development medicines for pulmonary fibrosis.
