SOCS7通过抑制细胞凋亡促进肺癌A549细胞增殖的机制研究

摘要 目的研究SOCS7对肺癌A549细胞增殖的影响及可能的作用机制。方法在肺癌A549细胞中分别采取过表达SOCS7和干扰SOCS7的方法,以细胞计数法(CCK-8)检测各实验组细胞的增殖情况;以Transwell方法检测各实验组细胞的迁移能力;以蛋白质印迹法(Western blot)检测各实验组凋亡相关基因的蛋白表达量情况;以ELISA方法检测各实验组免疫相关基因的蛋白表达情况。结果干扰SOCS7时,转染96 h后,与对照组和转染controlsiRNA组相比,肺癌A549细胞的增殖力明显降低,迁移能力明显降低。Western blot实验结果显示细胞中Bcl-2表达量显著降低,Cleaved Caspase3表达量升高;干扰SOCS7时,转染12 h后,与对照组和转染controlsiRNA组对比,肺癌A549细胞中人肿瘤坏死因子α(Tnf-α)和IL-1表达量显著升高,IL-6无显著性变化,在转染24 h后,与对照组和转染空载体组对比,IL-6表达量显著升高。结论干扰SOCS7能够抑制肺癌A549细胞的增殖和迁移能力,降低Bcl-2蛋白表达量,提高Caspase-3蛋白表达量,Tnf-α和IL-1表达量也显著升高,SOCS7可能通过炎症反应和Caspase-3途径来调节肺癌A549细胞的生长。 Objective To study the effect of SOCS7 on the proliferation of lung cancer A549 cells and its possible mechanism.Methods Over-expression of SOCS7 and interference with SOCS7 were used in lung cancer A549 cells.The cell count method(CCK-8)was used to detect the value-added status of cells in each experimental group;the Transwell method was used to detect the migration ability of cells in each experimental group;Western blot was used to detect the protein expression of apoptosis-related genes in each experimental group;ELISA was used to detect the protein expression of immune-related genes in each experimental group.Results When interfering with SOCS7,96 h after transfection,compared with the control group and the control siRNA transfection group,the proliferation ability of lung cancer A549 cells was significantly reduced,and the migration ability was significantly reduced.The expression of Bcl-2 in lung cancer A549 cells was significantly reduced,and the expression of Cleaved Caspase 3 was increased;when SOCS7 was interfered,12 h after transfection compared with the control group and the control siRNA transfection group.The expression levels of human tumor necrosis factorα(Tnf-α)and IL-1 in lung cancer A549 cells were significantly increased,and there was no significant change in IL-6.After 24 hours of transfection,compared with the control group and the transfected empty vector group,IL-6 expression increased significantly.Conclusion Interference with SOCS7 can inhibit the proliferation and migration of lung cancer A549 cells,reduce the expression of Bcl-2 protein,increase the expression of Caspase-3 protein,and the expression of Tnf-αand IL-1 also increase significantly.SOCS7 may the Caspase-3 pathway regulates the growth of lung cancer A549 cells.
