Expression of ALDH1 and AQP-5 proteins in NSCLC tissues and their relationship with clinical pathological parameters and prognosis of patients

摘要 目的观察ALDH1和AQP-5蛋白在非小细胞肺癌(NSCLC)组织中的表达,并分析与NSCLC临床病理学参数、预后的关系。方法收集2010年1月至2013年12月手术切除的160例经病理学证实为NSCLC患者的组织标本及其癌旁正常组织标本,免疫组化检测ALDH1和AQP-5的表达,并分析二者与NSCLC临床病理学参数、预后的关系。结果ALDH1、AQP-5蛋白主要表达于细胞核中,ALDH1在NSCLC组织和癌旁正常组织中阳性表达率分别为90.00%和25.00%,AQP-5在NSCLC组织和癌旁正常组织中阳性表达率分别为93.75%和17.50%,NSCLC组织中ALDH1和AQP-5阳性表达率明显高于癌旁组织,差异有统计学意义(P<0.001);ALDH1、AQP-5蛋白表达在腺癌和鳞癌间无统计学意义(P>0.05),而在分化程度、TNM分期和淋巴结是否转移间存在统计学意义(P<0.05);Spearman相关分析结果显示ALDH1、AQP-5蛋白在NSCLC组织呈明显的正相关关系(r=0.726,P<0.001);ALDH1和AQP-5表达强度为-/+的NSCLC患者中位生存期较表达强度为++/+++的患者明显延长,差异有统计学意义(P<0.05)。Cox单因素回归分析显示分化程度、TNM分期、淋巴结是否转移及ALDH1与AQP-5的表达是影响NSCLC临床预后的因素(P<0.05);而Cox多因素回归分析结果显示TNM分期、ALDH1和AQP-5表达是影响NSCLC预后的风险因素(P<0.05)。结论ALDH1和AQP-5过表达可促进NSCLC的发生发展,降低NSCLC患者的生存期,联合监测ALDH1和AQP-5的表达对NSCLC的发生发展有一定的临床价值。 Objective To investigate the expression of ALDH1 and AQP-5 proteins in non-small cell lung cancer(NSCLC)tissues,and to analyze the relationship between ALDH1,AQP-5 proteins and the clinical pathological parameters as well as the prognosis of patients with NSCLC.Methods A total 160 tissue specimens and adjacent normal tissue specimens from 160 patients with NSCLC who were pathologically confirmed and were resected in our hospital from January 2010 to December 2013 were collected.The expression levels of ALDH1 and AQP-5 were detected by immunohistochemistry,and their relationship with clinical pathological parameters and prognosis of patients with NSCLC was analyzed.Results ALDH1 protein and AQP-5 protein were mainly expressed in the nucleus,and the positive expression rate of ALDH1 in NSCLC tissues and in normal tissue adjacent to carcinoma was 90.00%and 25.00%,respectively,that of AQP-5 in NSCLC tissues and in normal tissue adjacent to carcinoma was 93.75%and 17.50%,respectively,and the positive expression rates of ALDH1 and AQP in NSCLC tissues were significantly higher than those in normal tissue adjacent to carcinoma(P<0.01).There were no significant differences in the expression levels of ALDH1 and AQP-5 proteins between adenocarcinoma and squamous cell carcinoma(P>0.05),however,there were significant differences in different differentiation degree,TNM stage and lymph node metastasis(P<0.05).Spearman correlation analysis showed that there was a significant positive correlation between ALDH1 and AQP-5 in NSCLC tissues(r=0.726,P<0.01).The median survival period of NSCLC patients with ALDH1 and AQP-5 expression intensity of-/+was significantly longer than those with expression intensity of++/+++(P<0.05).Cox univariate regression analysis showed that differentiation degree,TNM stage,lymph node metastasis,the expression levels of ALDH1 and AQP-5 were the risk factors affecting the clinical prognosis of patients with NSCLC(P<0.05).Cox multivariate regression analysis showed that TNM staging,ALDH1 and AQP-5 expression were the risk factors affecting the prognosis of patients with NSCLC(P<0.05).Conclusion The overexpression of ALDH1 and AQP-5 can promote the pathogenesis and development of NSCLC,and reduce the survival period of patients with NSCLC.The combination monitoring of the expression levels of ALDH1 and AQP-5 has certain clinical value in evaluating the occurrence and development of NSCLC.
