贵州山核桃转录组文库构建与测序 被引量:1

摘要 为探讨贵州山核桃(Carya cathayensis Sarg.)转录组功能基因信息,首次使用Illumina Hiseq 2500平台对贵州山核桃进行转录组测序分析,共获得43241968个reads片段。经序列组装共计获得98117条unigene,总长度高达80841463 bp,74.22%的unigene长度集中在1~1000 bp之间。功能注释结果显示,有58464个unigene在NR数据库获得注释,注释到葡萄科的占总的unigene 28.7%。另外对获得注释的序列信息进行初步分析,选取生物学特征重要基因(生长相关基因)和油脂合成相关基因进行初步筛查,分别获得15个和16个基因,填补了贵州山核桃转录组功能基因信息空白,为进一步开展贵州山核桃重要品质基因挖掘和功能标记开发奠定了重要的分子基础。 In this paper,the Illumina Hiseq 2500 platform was used for the first time to analyze the transcriptome functional gene information of Carya cathayensis Sarg.A total of 43241968 reads fragments were obtained from the transcriptome sequencing of C.cathayensis.A total of 98117 unigenes were obtained by sequence assembly,with a total length of 80841463 bp,and 74.22%of the unigenes were concentrated in the range of 1-1000 bp.The results of functional annotation showed that 58464 unigenes were annotated in the NR database,and 28.7%of the total unigenes were annotated to grape families.In addition,the annotated sequence information was preliminarily analyzed,and the important genes of biological characteristics(growth-related genes)and lipid synthesis-related genes were selected for preliminarily screening,and 15 and 16 genes were obtained,respectively.These studies filled the gap of functional gene information in the transcriptome of C.cathayensis and laid an important molecular foundation for further exploration of important quality genes and development of functional markers in C.cathayensis.
