摘要 目的:研究黄芪多糖(APS)对卵巢癌细胞顺铂(DDP)化疗的影响及其机制。方法:不同浓度APS联合DDP处理体外培养的卵巢癌SKOV3细胞,MTT法检测细胞增殖,流式细胞术检测细胞凋亡,PCR和Western Blot检测Caspase-3、Bax、Bcl-2基因及蛋白表达变化。结果:药物作用24 h时,随着APS浓度增加,SKOV3细胞增殖受到抑制,凋亡率逐渐增加,并呈浓度依赖性(均P<0.05)。药物作用时间为48 h、72 h时,APS对SKOV3细胞增殖有抑制作用,但不同浓度间抑制率无明显差异(P>0.05)。联合用药组的Bax、Caspase-3的蛋白和基因表达随着APS浓度的增高逐渐增加,Bcl-2的蛋白和基因表达逐渐减少(均P<0.05)。结论:APS有潜在抗卵巢癌作用,可提高卵巢癌细胞对DDP的敏感性,其作用机制可能是通过上调凋亡相关因子而促进卵巢癌细胞凋亡。 Objective:To study the effect and mechanism of astragalus polysaccharide(APS)on cisplatin(DDP)chemotherapy in ovarian cancer.Methods:Ovarian cancer SKOV3 cells were treated with APS combined with DDP at different concentrations.Cell proliferation and apoptosis were detected by MTT and flow cytometry,respectively.The mRNA and protein expression of Caspase-3,Bax,Bcl-2 were detected by RT-PCR and Western Blot.Results:With the increase of APS concentration,the proliferation of ovarian cancer SKOV3 cells was inhibited and the apoptosis rate increased gradually in a concentration-dependent manner at 24 h after drug treatment(all P<0.05).APS had inhibitory effect in SKOV3 cells proliferation,but there was no significant difference among different concentrations either at 48 h or 72 h after drug treatment(P>0.05).The protein and mRNA expression of Bax,Caspase-3 in SKOV3 cells were increased,while Bcl-2 were decreased gradually,with the increase of APS concentration(all P<0.05).Conclusion:APS have potential anti-ovarian cancer effect and could increase the sensitivity of ovarian cancer cells to DDP,which may be dependent on up-regulating apoptosis-related factors to promote apoptosis of ovarian cancer tumor cells.
